Transplantation of UC-MSCs on collagen scaffold activates follicles in dormant ovaries of POF patients with long history of infertility

Abstract: Premature ovarian failure (POF) is a refractory disease for clinical treatment with the goal of restoring fertility. In this study, umbilical cord mesenchymal stem cells on a collagen scaffold (collagen/UC-MSCs) can activate primordial follicles in vitro via phosphorylation of FOXO3a and FOXO1. Transplantation of collagen/UC-MSCs to the ovaries of POF patients rescued overall ovarian function, evidenced by elevated estradiol concentrations, improved follicular development, and increased number of antral follic… Show more

“…Other work has found that MenSCs produce functional spheroids, which appear to be essential for their observed therapeutic effects (86). FOXO3a (96,97). Furthermore, UC-MSCs have been reported to induce angiogenic growth factors, including VEGF, hepatocyte growth factor (HGF), placental growth factor (PGF), and transforming growth factor-beta 1 (TGF-β1) (12,98).…”

“…Finally, in the most recent interventional study, Ding et al (2018) used ovarian stem cell injection. Fourteen patients with idiopathic POI were randomly assigned to receive either ovarian collagen and umbilical cord mesenchymal stem cell (UC-MSC) ovarian transplantation plus HRT (premarin combined with provera) (group one), or UC-MSC only ovarian transplantation plus HRT (premarin combined with provera) (group two).…”

“…In group one, however, the pregnancy was terminated owing to trisomy 21; the patient was 37 years old. The other pregnancy, in group two, was ongoing at the time of publication; the patient was 34 years old (Ding et al, 2018). The small sample size of these studies is their main limitation.…”
