Transplantation of Human Umbilical Cord Blood-Derived Mesenchymal Stem Cells Improves Cartilage Repair in a Rabbit Model

Abstract: The aim of this study was to investigate the therapeutic efficacy and safety of transplanting human umbilical cord blood-derived mesenchymal stem cells (hUCB-MSCs) in the treatment of cartilage injury. First, the articular cartilage defect model in rabbits was constructed. Then, the identified hUCB-MSCs and rabbit bone marrow stem cells (rBM-MSCs) were transplanted into the bone defect, respectively, and the cartilage repair effect was observed by hematoxylin-eosin (HE) staining and immunohistochemistry. Besid… Show more

“…However, it is worth noting that the simulation of joint cartilage damage in numerous publications is carried out surgically, creating mechanical damage to the cartilage [46]. Therefore, some researchers, considering the prospects of using MSCs in acute cartilage injury, take into account the data obtained in such studies [43].…”

“…The surface of cartilage slices was evenly covered with DAB, stained with hematoxylin, and soaked in 1% hydrochloric acid alcohol after rinsing with distilled water for 5 min. Tissue samples were deparaffinized and rehydrated by using xylene substitute and graded alcohols [ 34 ]. The images were observed using a microscope (Olympus BX-50, Tokyo, Japan) with high magnification (×200).…”

“…In addition, Yan et al verified that the umbilical cord MSCs (UMSCs-EXO) exosomes had powerful cartilage tissue protection effects, containing activating the proliferation and migration of chondrocytes, enhancing matrix synthesis and impeding chondrocyte apoptosis [39]. In the constructed animal model of cartilage defect in vivo, after intra-articular injection of UMSCs-EXO, the cartilage defect was repaired well [40].…”
