Transmitted Antiretroviral Drug Resistance and Thumb Subdomain Polymorphisms Among Newly HIV Type 1 Diagnosed Patients Infected with CRF01_AE and CRF07_BC Virus in Guangdong Province, China

Abstract: The aim of this study was to elucidate the prevalence of transmitted drug-resistant (TDR) mutations and reverse transcriptase (RT) thumb subdomain polymorphisms in CRF01_AE and CRF07_BC virus among newly diagnosed, therapy-naive HIV-1 patients in Guangdong Province, China. One hundred and sixty-four samples were collected in the Guangzhou Eighth People's Hospital. The entire protease gene and 300 codons of the entry part of the reverse transcriptase were amplified and sequenced. Furthermore, genotypic drug res… Show more

“…1A). Similar to CRF01_AE cluster 1, none of the CRF07_BC sequences available in the database (n = 487) from non-MSM high-risk populations belonged to CRF07_BC cluster 3 except for a few CRF07_BC strains from Guangdong IDUs (6 of 35)32 (Fig. 1B).…”

“…A survey from the prefecture reveals that the prevalence of TDR was 4.3% after 7 years of the scale-up [19]. Another study from Guangdong province showed that the prevalence of TDR was 6.7% after 5 years of the scale-up [20]. A report from Henan province, the first province to provide free ARV to HIV-infected individuals in 2003 in China, shows that the prevalence of TDR in the province is 14.3%, 8 years after the scale-up [21].…”

“…Participants were identified with primary infection by screening 2000 MSM for HIV infection every 2 months using enzyme-linked immunosorbent assay for antibody testing and nested polymerase chain reaction for HIV RNA testing [2,7]. All participants were diagnosed with HIV within 2 months of their infection and had their blood collected for CD4 T-cell count, viral load measurements, and HIV genotyping [2,5,8]. Sequences covered a 1.3-kb region of the HIV-1 pol, including protease (amino acids 1-99) and partial reverse transcriptase (amino acids 1-305; GenBank accession numbers KP250654-KP250829).…”
