Transmission Failure in Sympathetic Nerves Produced by Hemicholinium

Abstract: It has been shown by others that hemicholinium (α,α′‐dimethylethanolamino‐4,4′‐biacetophenone) inhibits the synthesis of acetylcholine, an effect which is reversed by choline. Hemicholinium produces a failure of response to nerve stimulation in the following sympathetically innervated preparations: guinea‐pig isolated vas deferens, rabbit isolated uterus, rabbit isolated colon, perfused rabbit ear, cat isolated atria and the piloerector muscles in the cat's tail. The blocking action of hemicholinium on the res… Show more

“…Hemicholinium can reduce the release of acetylcholine from cholinergic nerve endings by interfering with the synthesis of acetylcholine in the rat phrenic nervediaphragm preparation (Reitzel & Long, 1959), in parasympathetic nerves in the rabbit atria (Chang & Rand, 1960) and in vagal nerve endings in the rabbit colon (Wong & Long, 1961). On the circular muscle strip preparation, hemicholinium abolished the effects of nicotine, 5-hydroxytryptamine and histamine, presumably through a similar mechanism of action.…”

“…Since hemicholinium is moderately active in blocking the hypogastric nerve (Chang & Rand, 1960) it was surprising to find that, in 5 experiments, this drug at levels of 1 x 10-4 had absolutely no effect on the response of the Finkleman preparation to stimulation of the periarterial nerves, even when allowed to act for periods of 45 min.…”

“…Isolated atria from young cats with their sympathetic nerves attached were prepared as described by Chang &Rand (1960); in addition the vagus nerves were attached (Burn & Rand, 1958a).…”
