Agrobacterium-mediated genetic transformation is the most widely used technology to obtain overexpression of recombinant proteins in plants. Molecular events that occur within Agrobacterium during interactions with host plants have been studied extensively, and now we have a reasonable understanding the key factors involved in the regulation of T-DNA nuclear import and genomic integration. By contrast, very little is known about the events that take place in the host cells during genetic transformation by Agrobacterium. Here, we describe the plant-related factors including genotype, genes, proteins, competency of target tissues and phenolic compounds that participate in Agrobacteriummediated genetic transformation and discuss their possible roles in this process. Because Agrobacterium probably adapts existing cellular processes for its life cycle, identifying the processes in host cells during Agrobacterium infection might contribute to better understanding of basic biological processes as cell communication, intracellular transport and DNA repair and recombination as well as to expanding the host range of Agrobacterium as a genetic engineering tool.

Agrobacterium-mediated transformation of European chestnut embryogenic cultures

An innovative and efficient genetic transformation protocol for European chestnut is described in which embryogenic cultures are used as the target material. When somatic embryos at the globular or early-torpedo stages were cocultured for 4 days with Agrobacterium tumefaciens strain EHA105 harbouring the pUbiGUSINT plasmid containing marker genes, a transformation efficiency of 25% was recorded. Murashige and Skoog culture medium containing 150 mg/l of kanamycin was used as the selection medium. The addition of acetosyringone was detrimental to the transformation efficiency. Transformation was confirmed by a histochemical beta-glucuronidase (GUS ) assay, PCR and Southern blot analyses for the uidA (GUS) and nptII (neomycin phosphotransferase II) genes. At present, 93 GUS-positive chestnut embryogenic lines are being maintained in culture. Low germination rates (6.3%) were recorded for the transformed somatic embryos. The presence of the transferred genes in leaves and shoots derived from the germinated embryos was also verified by the GUS assay and PCR analysis.

Tea is not only the most important nonalcoholic beverage drinks worldwide, but also an important “health drink”. It is served as morning drink for a two‐third of world populace daily. Although conventional breeding and propagation contributed significantly in the last several decades for varietal improvement, due to the limitations of conventional breeding, application of biotechnology becomes an alternative approach. Further, its long life cycle, self‐incompatibility, and high inbreeding depression limit its genetic improvement either via the selection of elites from existing natural populations or by the development of biclonal hybrids. With the era of genomics, several genes from tea and its wild genera have been cloned. Thus genetic transformation of tea is important to overcome the limitations of breeding with the assistance of genomics for increasing productivity with lower cost of production, a need of tea industry today. The present report deals with in‐depth progress of various aspects of genetic transformation from tea and related genera, which will be a valuable information for the workers working on various aspects of Camellia biotechnology.
