Transformation of patchouli alcohol to β-patchoulene by gastric juice: β-patchoulene is more effective in preventing ethanol-induced gastric injury

Abstract: Pogostemonis Herba is a functional food approved in Asian countries. Its major constituent, patchouli alcohol (PA), possesses a gastroprotective effect and is reported to transform into β-patchoulene (β-PAE) under acidic conditions. To investigate whether β-PAE, the metabolite of PA, has a protective effect on the gastrointestinal tract, the formation of β-PAE by gastric juice and the anti-ulcerogenic potential of β-PAE against ethanol-induced gastric injury were evaluated. The Results indicated that PA was co… Show more

“…Oxidative stress plays a significant role in the pathogenesis of gastric injury induced by ethanol (29). Ethanol administration causes the overproduction of reactive oxygen species (ROS) and acceleration of lipid peroxidation to aggravate gastric injury (30). Because ROS could convert unsaturated fatty acids to MDA through lipid peroxidation, MDA is usually used as a marker of lipid peroxidation (31).…”

“…The blood was immediately centrifuged at 3,500 rpm for 15 min at 4°C and the serum (supernatant) was collected according to the instructions of the manufacturer of nitric oxide (NO) and IL-6 (Nanjing Jiancheng Bioengineering Institute, Nanjing, China). The levels of NO and IL-6 in the serum were measured via the nitrate reductase and enzyme-linked immunosorbent assay method (Almasaudi et al, 2016; Liu et al, 2017).…”

“…Prior to experimentation, all rats were fasted for 24 h with access only to clean water and accommodated in uniform mesh cages to prevent coprophagy. One hour after the final administration, animals except normal group were orally given with absolute ethanol (0.5 mL/100 g body weight) to establish acute gastric damage model, as previously mentioned method [19]. Blood samples from each rat were collected for serum biochemistry tests after 60 min, then animals were euthanized, and their stomachs were rapidly removed for further analysis.…”
