Transfer of novel storage proteins from a synthetic hexaploid line into bread wheat

Abstract: Pure line selection for the presence of Glu-D1-4t+10.1t in high molecular weight glutenin subunits was applied in a cross between a synthetic hexaploid wheat (SHW) 530-1 (Triticum dicoccum / Aegilops tauschii acc. 19088) (2n = 42, . Grain number and seed weight per plant of selected BC 1 F 5-7 individuals accompanied this high molecular weight subunit pair to create 9 wheat genotypes. They were tested in the field and showed sufficient germination and winter survival with high seed set. Hierarchical cluster an… Show more

“…Ten grains from each line were crushed and ground to powder. Extraction of high molecular weight-glutenin subunits (HMW-GS) was performed in four stages (Singh et al, 1991) as described by Doneva et al (2018). The electrophoresis run on a vertical apparatus in two ways: 1) classical one-dimensional 12% polyacrylamide gel; 2) one-dimensional 12% polyacrylamide gel SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis) with the addition of 4М urea (Lafiandra et al, 1993).…”

“…Six wheat genotypes in BC 1 F 9 generation (38-7, 39a-5, 40-3, 41a-6, 45-5, and 46-7) among the nine selected introgression lines (Doneva et al, 2018) Sowing and harvesting were done by machine and grain yield was calculated in t/ha. Disinfection of seeds with the commercial product RAXIL 060 FS (tebuconazole 60 g/l), applied in a standard dose of 50 ml/100 kg of seeds for semi-wet disinfection, consumable working solution 2 l/100 kg of seeds; loading date -there is one treatment with an insecticide (commercial product DECA EC, deltamethrin) against enemies and a fungicide for rust and powdery mildew species (product AMISTAR 25 SK, azoxystrubin).…”

“…Evaluation of wheat baking quality involves numerous tests of traits such as protein concentration, gluten index, hardness index, water absorption, sedimentation value, falling number, etc. (Peña et al, 2002; Kurt-Polat, Yagdi, 2017; Doneva et al, 2018). Baking quality of wheat is mostly determined by protein quantity and quality grading.…”
