Film-forming sprays offer many advantages compared to conventional topical preparations because they can provide uniform drug distribution and dose, increased bioavailability, lower incidence of irritation, continuous drug release, and accelerated wound healing through moisture control. Film-forming sprays consist of polymers and excipients that improve the characteristics of preparations and enhance the stability of active substances. Each type of polymer and excipient will produce films with different features. Therefore, the various types of polymers and excipients and their evaluation standards need to be examined for the development of a more optimal form of film-forming spray. The selected literature included research on polymers as film-forming matrices and the application of these sprays for medical purposes or for potential medical use. This article discusses the types and concentrations of polymers and excipients, sprayer types, evaluations, and critical parameters in determining the sprayability and film characteristics. The review concludes that both natural and synthetic polymers that have in situ film or viscoelastic properties can be used to optimise topical drug delivery.

The medicine was given to the target site with a pressurized topical spray (aerosol pharmaceutical) and a non-pressurized topical spray solution by gently pressing the spray container against the skin and initiating the spray. Formulation components consists of product concentrate (one or more pharmaceutical drugs with relevant excipient) and packaging components (valves, pump, actuator, container). After formulation process development various quality control test like spray pattern, spray content uniformity and geometry, particulate matter, droplet size distribution, microbial limit, net content, pH, osmolarity, weight loss, leachability, viscosity, valves, actuators, diptubes, weight checking, leak test, valve discharge rate, dosage with valves, therapeutic activity, and toxicity studies to check the quality and efficacy. The benefit of Topical Spray Life Cycle Management (post-approval adjustments, 30-day supplements, annual report, Food and Drug Administration inspection and enforcement, cover letter, risk management standard and related information from the International Organization for Standardization) is that it covers the entire lifecycle of a topical spray, from conception through formulation design development to marketing. The product lifecycle management data offers regulatory bodies with a significant financial benefit. Topical non-pressurized sprays have been developed for a variety of applications due to their ease of use, environmental friendliness, low cost, and innovative technology. This review give details about meter dose topical spray of Estradiol, 1% tolnaftate, 2% Miconazole nitrate spray, 0.5% w/w Voriconazole spray, Clotrimazole transdermal apray, Lopinavir meter dose transdermal spray are studied by researcher. This review also enlisted patents and marketed formulation on topical spray from year 2007 to 2020.

Liquid spray preparations for cutaneous administration are solutions, suspensions or emulsions that use compressed or converted gas to a liquid state, to disperse the active substance in the form of aerosols or they are propellant-free (not pressurized) pumps and can also disperse the active substance in the form of aerosols. The paper presents data from professional regulations and relevant literature related to dosage forms, characteristics and composition of cutaneous sprays, and on inner packaging (containers and pump/valve systems and sprays), which is their main specificity. They are most commonly formulated as cutaneous spray, solution, for local administration of antimicrobials, or for regional delivery of nonsteroidal anti-inflammatory drugs (NSAIDs) and transdermal delivery of hormones. In the development of transdermal sprays, great attention is paid to strategies for improving the drug delivery and therapeutic efficacy (e.g., by using permeation enhancers, colloidal carriers). Equally important is design of inner packaging that should ensure preservation of drug quality, but also precise and reproductive dosing according to the individual therapeutic needs of patients, which, with simple application, good tolerance and satisfactory aesthetic properties, is a significant advantage of cutaneous sprays compared to other liquid and semisolid preparations for cutaneous administration as well as transdermal patches.
