Transcriptional regulation of plant senescence: from functional genomics to systems biology

Abstract: Leaf senescence is an active process that involves the increased expression of many hundreds of genes. Many putative transcription factors show enhanced transcription during leaf senescence in Arabidopsis and functional analysis of these should help to indicate their role in controlling gene expression during leaf senescence. In this paper, we describe the analysis of knockout insertion mutants in two different senescence-enhanced genes, one encodes a heat shock transcription factor and the other a zinc finger… Show more

“…In this analysis, we identified differentially expressed TFs mainly from the NAC, MYB, AP2-EREBP, HB and bZIP families ( Figure 5). Members of these TF families have been widely studied and reported as candidate genes activating the leaf senescence programme in different species (Balazadeh et al, 2008;Breeze et al, 2008). Genes with high sequence similarity to ANAC055, ANAC019 and ANAC029 from A. thaliana were highly expressed during sunflower senescence (AT3G15500, AT1G52890 and AT1G69490, respectively) (Table S3).…”

“…Other biochemical, cellular, integrative and adaptive systems became accreted on this armature as the evolving plant encountered new environmental and developmental contexts. Current systems biology approaches [ 184 ] are leading towards the idea of senescence as a complex of reticulated gene interactions with nodes and hubs and away from the older conception of a master switch from which subsequent events propagated [ 5 ]. The evolutionary perspective is entirely consistent with such a system network model of plant senescence and its control.…”

“…Regulation by ANAC092 of several other TFs known to be stress related is also predicted in this network model. ANAC019, ANAC055, and RD26 (ANAC072) have all been shown to have a role in drought stress ; MYB2 has a role in ABA signaling and salt stress (Abe et al, 2003;Yoo et al, 2005), and PMZ, a zinc finger protein, has a role in stress-induced senescence (Breeze et al, 2008). The gene regulatory network model also predicts feedback and feed-forward connections between ANAC092, STZ, and TBP1-1, which encodes a telomere binding protein.…”
