Transcriptional mechanisms regulating Ca2+ homeostasis

Abstract: Ca2+ is a dynamic cellular secondary messenger which mediates a vast array of cellular responses. Control over these processes is achieved via an extensive combination of pumps and channels which regulate the concentration of Ca2+ within not only the cytosol but also all intracellular compartments. Precisely how these pumps and channels are regulated is only partially understood, however, recent investigations have identified members of the Early Growth Response (EGR) family of zinc finger transcription factor… Show more

“…Furthermore, Ca 2ϩ oscillations regulate SERCA2 expression in astrocytes after growth factor receptor stimulation (65) and in human lens epithelial cells (66), although there is no evidence of NFAT binding to the SERCA2 promoter. In addition, MAPK-and PKC-dependent induction of Erg1 results in down-regulation of the SERCA2 gene, although, again, the effect is probably indirect because no functional Erg1-binding sites are found in the SERCA2 promoter (10,67,68).…”

“…Of the different control mechanisms, here we will review those that control Ca 2ϩ homeostasis at the transcriptional level and that are directly regulated by Ca 2ϩ . A review of the transcriptional control of calcium homeostasis, with particular emphasis on the roles of members of the Egr (early growth response) family of zinc finger immediate-early transcription factors and the closely related protein WT1 (Wilms tumor suppressor 1), has been published recently (10).…”

“…Furthermore, Egr-1 deficiency is very sensitive to the perturbation of intracellular calcium homeostasis, because only treatment of Egr-1 knockdown cells with thapsigargin, which inhibits SERCA leading to calcium depletion in ER, causes further increases in ER stress and cell death. Altered calcium regulation has been found in Egr1 −/− cardiomyocytes through downregulation of major proteins involved in control of calcium homeostasis [35,36]. Thus, we speculate that attenuation of ER stress by Egr-1 in β-cells may mediate through remodeling of calcium homeostasis.…”

“…Існує [92]. Беру-чи до уваги, що зміна експресії Са 2+ -помпи ПМ відбувається за багатьох захворювань (діабет, пухлини, остеопороз) дослідження регуляції транскрипції гена цієї помпи потребує більшої уваги.…”
