Transcriptional factor specificity protein 1 (SP1) promotes the proliferation of glioma cells by up-regulating midkine (MDK)

Abstract: Midkine (MDK) expression is associated with the proliferation of many cancers, including glioma. SP1 directly up-regulates the expression of MDK, and the SP1-MDK axis cooperates in glioma tumorigenesis.

“…Nucleolin has been previously described as an ITAF for virus IRESs such as poliovirus (20). Recently, it has been found to enhance cellular IRES-dependent translation of specificity protein-1 (Sp1), a transcription factor involved in tumor cells proliferation (22,39). These results are in accordance with findings showing that IRES-dependent translation initiation is regulated by ITAF's subcellular location (19).…”

“…The [3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium] MTS viability assay (Promega) was performed according to standard methods as described previously (Luo et al, 2015). ZKSCAN1 and circZKSCAN1 knockdown or overexpression cell lines and control cells were seeded at 2 9 10 4 cells in 100 lL medium per well of a 96well plate Corning Incorporated (Corning, NY, USA) for the MTS assay (Qi et al, 2014;Qin et al, 2016).…”

“…In all networks, the most connected TF targeted by the studied miRNAs was Sp1 with 12, 32, and 32 connections in the PPI, hESCT0, and fBrain networks, respectively—Figure and Supporting Information Figure S5. Sp1 has been described as a promoter of proliferation and stem cell marker in glioma cells but it is still poorly characterized in neurogenesis.…”
