Transcription factor AhR regulates glutathione S-transferases (GSTs) conferring resistance tolambda-cyhalothrin inCydia pomonella

Abstract: Transcription factor aryl hydrocarbon receptor (AhR) can enhance insect resistance to insecticides by regulating the detoxification metabolic network. Our previous studies have confirmed that overexpression of cytochrome P450 monooxygenases (P450s) and glutathione S-transferases (GSTs) are both involved in lambda-cyhalothrin resistance in Cydia pomonella. In this study, we report that AhR regulates GSTs thus conferring lambda-cyhalothrin resistance in C. pomonella. Spatiotemporal expression patterns indicated … Show more

“…For instance, transcription factors CncC/Maf, CREB, HNF4, and AhR/ARNT have been proven to participate in the regulation of P450 genes mediating imidacloprid susceptibility in insects. 10,11,15,17 AP-1 protein was shown to mediate clothianidin resistance by regulating CYP6ER1 in N. lugens. 13 In the present study, it was found that FoxO mediated nitenpyram resistance in N. lugens, which might be through regulating CYP4CE1 expression by FoxO.…”

“…14 AhR regulated the expression of GSTe3, CYP9A121, and CYP9A122, thus conferring lambda-cyhalothrin resistance in Cydia pomonella. 15 AhR and ARNT in N. lugens were also reported to be involved in the susceptibility of imidacloprid, etofenprox, and isoprocarb. 16 NR members have also been implicated in the regulation of detoxifying enzyme genes involved in the metabolism of insecticides.…”

“…Following the injection of dsCpGPCR70, an assessment was conducted on the expression levels of detoxification genes, specifically selected as potential candidate genes known to play a role in the metabolic of lambdacyhalothrin in C. pomonella. 7 The result showed a notable decrease in mRNA levels of CpGSTs2 (Figure 6A) and CpCYP9A121 (Figure 6B) subsequent to the interference caused by CpGPCR70, in comparison with the control group. The enzymatic assay showed that the P450 enzyme activity decreased significantly at 12 h after the injection of dsCpGPCR70 (Figure 6D), while there was no significant difference of the GST enzyme activity compared with the control (Figure 6C).…”
