Recent studies suggested that Traditional Chinese Medicine could play a beneficial role in conventional cancer treatment. The aim of this retrospective cohort study was to investigate the effect of Chinese herbal medicine (CHM) combined with Western medicine on the survival of patients with colorectal cancer. A retrospective cohort study was conducted on patients with newly diagnosed colorectal cancer identified from the Dalin Tzu Chi Hospital Cancer Registry Database in 2004–2014. Combining with the medical records of the study hospital, patients were classified into CHM users and CHM nonusers. Kaplan–Meier analyses and Cox proportional hazards regression analyses were used to investigate the survival between CHM users and CHM nonusers. A total of 535 patients with colorectal cancer were included in the study with 147 of them were CHM users. The log-rank test for Kaplan–Meier survival curve revealed a statistically significant difference between the survival of CHM users and CHM nonusers (P=0.006). Results from multivariate Cox regression analysis showed that CHM use was significantly associated with better survival (adjusted hazard ratio = 0.54, 95% CI = 0.38 to 0.77). In addition, the CHM formulae Jia Wei Xiao Yao San, Zhi Bah Di Huang Wan, Ping Wei San, and Qui Pi Tang were significantly associated with better survival. In conclusion, findings from this retrospective cohort study indicated that integrated CHM and Western medicine could improve survival in patients with colorectal cancer. Additional research on integrating TCM with Western medicine to improve cancer survival is warranted.

Background: Recent findings suggested that premetastatic niche (PMN) is a prerequisite in mediating cancer metastasis. Previously we demonstrated that XIAOPI formula could inhibit breast cancer lung metastasis via inhibiting tumor associated macrophages (TAMs)-secreted CXCL1. Herein, we aimed to explore the effects of XIAOPI formula on preventing breast cancer PMN formation and its underlying molecular mechanisms.Methods: CXCL1 expression of TAMs was detected by qPCR and Western blotting assay. The influences of XIAOPI formula on the proliferation of TAMs and 4 T1 in the co-culture system were tested by CCK8 or EdU staining. Transwell experiment was applied to determine the effects of XIAOPI formula on the invasion ability of HSPCs and 4 T1. Breast cancer xenografts were built by inoculating 4 T1 cells into the mammary pads of Balb/c mice and lung metastasis was monitored by luciferase imaging. Immune fluorescence assay was used to test the epithelialmesenchymal transition process and PMN formation in the lung tissues. The effects of XIAOPI formula on TAMs phenotype, hematopoietic stem/progenitor cells (HSPCs) and myeloid-derived suppressor cells (MDSCs) were determined by flow cytometry.(Continued on next page)

Background: Colorectal cancer is a common malignant tumor and ranks third in cancer-related deaths. Considering that the side-effects of current therapies limit the clinical effectiveness, exploring new anti-colorectal cancer drugs from natural products is critical for the treatment of colorectal cancer. Among these drugs, liquiritin is an active component of the traditional Chinese herb Glycyrrhiza Radix and has been found to possess powerful anti-inflammatory and anti-tumor abilities. However, the direct molecular target of liquiritin remains unknown. Therefore, the aim of the present study was to identify potential molecular target of liquiritin to mediate its anti-colorectal cancer effect. Methods: The function of liquiritin in cell proliferation, apoptosis, migration and invasion was estimated by CCK-8 assay, colony formation, EdU assay, flow cytometry analysis, TUNEL assay, wound healing assay and transwell assay, respectively. Animal experiment was carried out to further confirm the role of liquiritin in vivo. H&E staining analysis, TUNEL staining and immunohistochemistry (IHC) assay were adopted for histological analysis. The mechanism research was conducted with RT-qPCR, western blot and luciferase report assay. Results: In this study, we found that liquiritin significantly inhibited the proliferation, migration, invasion and EMT processes of SW480 and HT-29 cells in a dose-dependent manner. MiRNAs have been extensively identified as drug targets in various studies. However, the miRNAs functioning as the direct targets of liquiritin remain unknown. In our study, we tested 8 potential pathogenic miRNAs screened from colorectal cancer patients, and we found that only the expression level of miR-671 was diminished due to liquiritin treatment. More importantly, overexpression of miR-671 partially abrogated the anti-tumor effects of liquiritin on colorectal cancer. Conclusions: Collectively, our findings demonstrated that liquiritin exhibits great potential in the treatment of colorectal cancer through regulation of miR-671.
