Tracking of transplanted human umbilical cord-derived mesenchymal stem cells labeled with fluorescent probe in a mouse model of acute lung injury

Abstract: The aim of the present study was topreliminarily visualize the distribution of humanumbilical cord-derived-mesenchymal stem cells (hUC-MSCs) in treating acute lung injury (ALI) using a targeted fluorescent technique. Anovel fluorescent molecule probe was first synthesized via the specific binding of antigen and antibody in vitro to label the hUC-MSCs. Two groups of mice, comprising a normal saline (NS)+MSC group and lipopolysaccharide (LPS)+MSC group, were subjected to optical imaging. At 4 h following ALI mou… Show more

“…Thus, it appears that very small amounts of transplanted cells have been introduced into the ovaries. On day 3, the SRY gene detected in ovarian tissue was not detected on day 7, which is consistent with studies showing a sharp decrease in fluorescence expression experiments on day 7 [26]. Similarly, only a very small amount of Prussian blue traces for Molday Iron were stained throughout the whole ovarian tissue, which appears to be some cells remaining after transplantation.…”

“…Meanwhile, the effect of hESC-MSC transplantation in cisplatin-treated mice was observed immediately after transplantation. Compared to the PBS group, rapid movement and atrophy of the body due to anticancer drugs have been restored, which is thought to be the result of the transplanted cells reaching each organ damaged by the MSC homing effect [26].…”

“…Current treatments focus on support, as mechanical ventilation is a cornerstone life-saving treatment for ARDS. Lung protective ventilation acts by limiting the iatrogenic injury that is linked to mechanical ventilation [2, 3] and is frequently used ventilation method. It includes several components, the most important of which is lowering tidal volume (V T ), limiting plateau (Pplat) to or below 30 cmH 2 O and higher positive end-expiratory pressures (PEEPs).…”

“…The lung showed strong fluorescence signals immediately after MSC injection as the majority of administrated cells were initially trapped inside lung capillaries. 40 Later, the circulating MSCs gradually accumulated in the liver and spleen. More importantly, the real-time traffic of single-cell clusters at several positions in the circulation system was observed, as well as their migration through blood vessels from one organ to another (Video S1 and S2).…”
