TPGS‐Functionalized Polydopamine‐Modified Mesoporous Silica as Drug Nanocarriers for Enhanced Lung Cancer Chemotherapy against Multidrug Resistance

Abstract: A nanocarrier system of d-a-tocopheryl polyethylene glycol 1000 succinate (TPGS)-functionalized polydopamine-coated mesoporous silica nanoparticles (NPs) is developed for sustainable and pH-responsive delivery of doxorubicin (DOX) as a model drug for the treatment of drug-resistant nonsmall cell lung cancer. Such nanoparticles are of desired particle size, drug loading, and drug release profile. The surface morphology, surface charge, and surface chemical properties are also successfully characterized by a ser… Show more

“…Considering the NIR photothermal responsiveness of the PDA‐based materials, the photothermal generating ability of the multifunctional nanoparticles was then investigated by real‐time detecting temperature changes of the nanoparticle in water solutions under 808 nm NIR laser irradiation . As shown in Figure a,e, after 5 min of irradiation under 808 nm laser (1 W cm −2 ), the temperature of the PDA@CP and PDA@CP‐PEG solutions (0.25 mg mL −1 ) could rapidly increase from 25 °C up to 49.1 and 45.6 °C, respectively.…”

“…Although chemotherapy, as a major cancer treatment approach, has greatly improved the survival rate, its clinical performance is still restricted due to systemic toxicity and lack of specificity . The design and preparation of prodrugs have been a popular strategy to overcome these drawbacks of chemotherapeutic drugs .…”

“…[5,6] Although chemotherapy,a samajor cancer treatment approach, has greatly improved the survival rate,i ts clinical performance is still restricted due to systemic toxicity and lack of specificity. [7,8] Thedesign and preparation of prodrugs have been ap opular strategy to overcome these drawbacks of chemotherapeutic drugs. [9] Prodrugs are derivatives of chemotherapeutic drugs with ab lockage of their active site, which exhibit low toxicity in normal tissues,a nd can be converted to their active, highly toxic forms in tumor tissues, triggered by the tumor microenvironment or external stimuli.…”

“…113 Cheng et al designed Datocopheryl polyethylene glycol 1000 succinate-functionalized polydopamine-modified MSNs loaded with DOX, and through the mice model tests proved that they had a strong inhibitory effect on MDR non-small-cell lung cancer cells, which provided a creative idea for the application of nanodrug delivery system to overcome the MDR. 114 …”
