Toxicity of thallium on isolated rat liver mitochondria: The role of oxidative stress and MPT pore opening

Abstract: Thallium(I) is a highly toxic heavy metal; however, up to now, its mechanisms are poorly understood. The authors' previous studies showed that this compound could induce reactive oxygen species (ROS) formation, reduced glutathione (GSH) oxidation, membrane lipid peroxidation, and mitochondrial membrane potential (MMP) collapse in isolated rat hepatocyte. Because the liver is the storage site of thallium, it seems that the liver mitochondria are one of the important targets for hepatotoxicity. In this investiga… Show more

“…In the present study, we used a rhodamine 123 exclusion assay to detect the membrane potential of cells due to its selective accumulation and quenching in mitochondria. Loss of the membrane potential will lead to release of the dye and, consequently, the fluorescence intensity [29]. Our data were shown as the percentage of MMP collapse ( %ΔΨm) in all treated groups.…”

“…In fact, our present study is in line to previous studies due to strong elevation in ROS production and attack to biological cell membrane caused to LPO. Also, experimental and clinical studies have shown that important site of ROS formation is related to mitochondria …”

“…Thallium binds to cysteine residues, preventing crosslinking bond formation critical to the keratinization of hair, a possible mechanism for the alopecia associated with acute thallium poisoning [7]. Thallium disrupts mitochondria [8] and interferes with energy production [9], including glycolysis, the Krebs cycle, and oxidative phosphorylation [9,10].…”
