Rhizoxin (1) ist ein 16-gliedriges Makrolid, das erstmals 1984 von Okuda und Mitarbeitern aus dem pflanzenpathogenen Pilz Rhizopus chinensis isoliert wurde und das als Phytotoxin die Reisfäule auslçst. [1,2] Hertweck und Partida-Martinez zeigten 2005, dass es sich bei 1 nicht um einen Pilzmetaboliten handelt -vielmehr wird es von einem bakteriellen Endosymbionten der Gattung Burkholderia produziert. [3] Rhizoxin (1) ist ein potenter Hemmer der eukaryotischen Tubulinpolymerisation [4] und hat sowohl in vitro wie auch in vivo eine deutliche Antitumoraktivität; [5] die Substanz wurde bis hin zur Phase II klinisch entwickelt, zeigte dort aber keine therapierelevante Wirkung. [6] Außer 1 selbst konnten aus Fermentationsbrühen von Rhizopus chinensis [4, 5a, 7] oder Burkholderia rhizoxina noch weitere verwandte Verbindungen isoliert werden, [8] darunter auch die Desepoxyvarianten WF-1360F (2) und Rhizoxin D (3); [4] für 2 wurde gegenüber 1 eine ähnliche [5a] oder sogar wesentliche stärkere zellwachstumshemmende Wirkung beschrieben. [8] Für Rhizoxine ist eine ganze Reihe von Totalsynthesen bekannt, von denen die meisten die Synthese von Rhizoxin D (3) zum Ziel hatten. [9] Bisher wurde erst eine einzige Synthese von 1 erfolgreich abgeschlossen, [10] und bis zum heutigen Tag wurde keine Synthese von 2 beschrieben. Wir waren nun im Rahmen einer geplanten Studie zur biologischen Aktivität seitenkettenmodifizierter Rhizoxine daran interessiert, eine neue Synthese von 2 und 3 zu entwickeln. Dabei wollten wir vor allem untersuchen, ob sich der Makrolactonring durch eine Ringschlussmetathese (RCM) [11] würde aufbauen lassen, ein Ansatz, der bei keiner der bekannten Rhizoxinsynthesen verfolgt worden war; als direktes Produkt der Cyclisierungsreaktion wurde dabei das Vinyliodid 5 anvisiert, das als gemeinsame Vorstufe für 2 und 3 wie auch für unterschiedlich seitenkettenmodifizierte Analoga fungieren sollte (Schema 1). Trotz umfangreicher experimenteller Versuche gelang es uns jedoch nicht, das Vinyliodid 5 über eine Ringschlussmetathese herzustellen, und zwar weder aus dem Dien 4 noch aus einer Vielzahl alternativer Substrate für die RCM-Reaktion (Schema 1). [12] Die ursprünglich vorgesehene Synthesestrategie musste somit modifiziert werden, und eine Mçglichkeit, die wir hierbei in Betracht zogen, war der Aufbau des Makrolactonrings durch eine Ringschluss-Alkinmetathese (RCAM). [13] Dies hätte es uns ermçglicht, die im Rahmen der Synthese von 4 gewonnenen Erkenntnisse (sowie entsprechende Zwischenprodukte) soweit wie mçglich in den neuen Syntheseansatz mit einzubeziehen. Die RCAM hat sich in jüngerer Zeit, und das vor allem wegen der Arbeiten von Fürstner und Mitarbeitern zur Entwicklung neuer Katalysatoren, [14] zu einem wertvollen Hilfsmittel für die Totalsynthese makrocyclischer Naturstoffe entwickelt. [15] Hier beschreiben wir nun die erste Totalsynthese des Naturstoffs WF-1360F (2), und zwar basierend auf einer RCAM-vermittelten Makrocyclisierung und der sich daran anschließenden, hochselektiven Umwandlung der Dreifachbin...
