Total polysaccharides of adlay bran (Coix lachryma-jobiL.) improve TNF-α induced epithelial barrier dysfunction in Caco-2 cellsviainhibition of the inflammatory response

“…Job's-tears (Coix lacryma-jobi L.) is an herbal plant species (Poaceae), native from Southeast Asia and used in traditional Chinese medicine. Besides its use in medicine due to the bioactive components, C. lacrymal-jobi arose as a potential staple crop to improve food security as an alternative to rice (Ruminta and Wicaksono 2017), due to high content of polysaccharides, proteins, lipids, polyphenols and phytosterols in the seeds (Li et al 2019). In addition, this plant can be used for alcoholic beverages (Wang et al 2012;Corke et al 2016).…”

“…Therefore, the development of effective and safe therapeutic agents for inflammatory bowel diseases is urgently needed. A growing body of research suggests that natural produces derived from edible fruit and food, such as flavonoids and polysaccharides, are potential agents for inflammatory bowel diseases treatment [16,24,25].…”

“…An increasing number of studies indicate that excessive production of inflammatory cytokines induces intestinal epithelial barrier dysfunction and eventually results in impairment of TJs protein integrity [24,29]. Additionally, the imbalance of proinflammatory factors, including IL-6, IL-8, and IL-1b, is involved in the progression of the gut inflammatory cascade [24,30]. TJs proteins play an important role in preserving intestinal epithelial barrier integrity by effectively sealing the paracellular pathway of Caco-2 cells [31].…”

“…It is not only a commonly used TCM, but it is also a commonly consumed food. Coicis semen has been proven to have numerous functions, such as detoxification and dampness and arthralgia removal; it also reduces cancer risk ( Yu et al, 2011 ; Bai et al, 2018 ; Duan, 2018 ; Xu et al, 2018 ; Choi et al, 2019 ; Huang Q. et al, 2019 ; Huang Y. Moreover, it has a wide range of anti-inflammatory, antioxidation, analgesic, and sedative effects, as well as pharmacological effects against gastric cancer, hepatocellular cancer, Lewis lung cancer, non-small cell lung cancer, pancreatic cancer, and pulmonary cancer (“Fuzheng” category represented by coicis semen oil) ( Manosroi et al, 2016a ; Manosroi et al, 2016b ; Qian et al, 2016 ; Xi et al, 2016 ; Wang et al, 2016 ; Han et al, 2017 ; Qu et al, 2017a ; Schwartzberg et al, 2017 ; Jang et al, 2018 ; Kim et al, 2018 ; Zhang et al, 2018 ).…”
