Topical TWEAK Accelerates Healing of Experimental Burn Wounds in Mice

Abstract: The interaction of tumor necrosis factor-like weak inducer of apoptosis (TWEAK) and its receptor fibroblast growth factor inducible 14 (Fn14) participates in inflammatory responses, fibrosis, and tissue remodeling, which are central in the repair processes of wounds. Fn14 is expressed in main skin cells including dermal fibroblasts. This study was designed to explore the therapeutic effect of TWEAK on experimental burn wounds and the relevant mechanism underlying such function. Third-degree burns were introduc… Show more

“…The mice were numbered and randomly assigned to either burn (3, 6, and 12 h) groups, treatment (0, 1, and 3 h) groups, or control group. Mice in both burn and treatment groups were anesthetized with 1% pentobarbital sodium and third-degree scald wounds were inflicted on the back as previously described ( 41 43 ). Briefly, a brass probe (diameter = 2 cm) was electrically preheated to 110°C and applied to the dorsal skin of the mice for 10 s. Mice in control group were only anesthetized with 1% pentobarbital sodium.…”

“…The burn site is inundated with inflammatory mediators and immune cells that exacerbate local tissue damage and further delay wound closure [461,462,463]. Inhibition of several cytokines has been shown to promote healing in burns [464,465].…”

“…TWEAK/Fn14 signaling modulates cutaneous inflammatory responses via regulating the cell cycle and cytokine secretion of keratinocytes as well as recruiting inflammatory cells to wound regions [20]. Topical application of recombinant TWEAK strengthens inflammatory cell infiltration and growth factor production, and it increases extracellular matrix components in wound areas [21].…”
