Toll‐Like Receptor 2–Deficient Mice Are Highly Susceptible toStreptococcus pneumoniaeMeningitis because of Reduced Bacterial Clearing and Enhanced Inflammation

Abstract: Toll-like receptor-2 (TLR2) mediates host responses to gram-positive bacterial wall components. TLR2 function was investigated in a murine Streptococcus pneumoniae meningitis model in wild-type (wt) and TLR2-deficient (TLR2(-/-)) mice. TLR2(-/-) mice showed earlier time of death than wt mice (P<.02). Plasma interleukin-6 levels and bacterial numbers in blood and peripheral organs were similar for both strains. With ceftriaxone therapy, none of the wt but 27% of the TLR2(-/-) mice died (P<.04). Beyond 3 hours a… Show more

“…Two recent reports investigated the role of TLR2 in pneumococcal meningitis in vivo and demonstrated an aggravated course in mice lacking TLR2 using a clinical severity score (20,21). One study provided survival data showing a 100% fatality rate in both TLR2 Ϫ/Ϫ and wt mice (21).…”

“…One study provided survival data showing a 100% fatality rate in both TLR2 Ϫ/Ϫ and wt mice (21). In both investigations, in which S. pneumoniae was injected intracisternally, differences between TLR2 Ϫ/Ϫ and wt mice in bacterial clearance were modest at best.…”

“…When compared with the WT mice the TLR2 knockout mice had greater bacterial loads in the brain, indicating that TLR2 conferred protective immunity against streptococcus pneumoniae infection in the CNS [103,104].…”
