Objectives:To estimate the fraction of head and neck cancer (HNC) attributable to tobacco and alcohol in cities in the Midwest, Southeast and South regions of Brazil. Methods: Case-control study including 1,594 cases of HNC and 1,292 hospital controls. The association of HNC with tobacco and alcohol was estimated by the odds ratio and respective 95% confidence intervals through non-conditional logistic regression, adjusted for age, sex, schooling, consumption of fruits and vegetables, alcohol drinking (to examine the tobacco effect), and tobacco smoking (to examine the alcohol effect). The proportions of HNC attributable to tobacco and alcohol were estimated through the attributable fraction (AF) calculation. Separate estimates were made for Goiânia (Midwest), Rio de Janeiro and São Paulo (Southeast) and Pelotas and Porto Alegre (South). Results: The HNC fraction attributable to smoking was slightly higher in Goiânia (AF = 90%) than in cities in the Southeast (AF = 87%) and South (AF = 86%). The HNC fraction attributable to the consumption of alcoholic beverages presented similar and higher results in the cities of Southeast (AF = 78%) and South (AF = 77%) than in Goiânia (AF = 62%). Conclusion: The HNC fractions attributable to smoking were more expressive than for alcohol consumption. Although with discrete distinctions between them, the AFs to tobacco smoking and alcohol consumption in HNC observed in the cities of these three Brazilian regions were similar to those obtained in Latin America studies, but they were higher than in other parts in the world.

Abstract. The aim of the present study was to investigate licochalcone-E (Lico-E)-induced apoptosis and the associated apoptotic signaling pathway in FaDu cells, a human pharyngeal squamous carcinoma cell line. Treatment with Lico-E exhibited significant cytotoxicity on FaDu cells in a concentration-dependent manner. The IC 50 value of Lico-E in FaDu cells was ~50 µM. Treatment with Lico-E increased the number of dead FaDu cells. Furthermore, chromatin condensation, which is associated with apoptotic cell death, was observed in FaDu cells treated with Lico-E for 24 h. By contrast, Lico-E did not produce cytotoxicity or increase the number of dead cells when applied to human normal oral keratinocytes (hNOKs). Furthermore, chromatin condensation was not observed in hNOKs treated with Lico-E. Treatment with Lico-E increased the expression of Fas ligand and the cleaved form of caspase-8 in FaDu cells. Furthermore, treatment with Lico-E increased the expression of pro-apoptotic factors, including apoptosis regulator BAX, Bcl-2-associated agonist of cell death, apoptotic protease-activating factor 1, caspase-9 and tumor suppressor p53, while decreasing the expression of anti-apoptotic factors, including apoptosis regulator Bcl-2 and Bcl-2-like protein 1 in FaDu cells. The expression of cleaved caspases-3 and poly (ADP-ribose) polymerase was significantly upregulated following treatment with Lico-E in FaDu cells, while Lico-E-induced apoptotic FaDu cell death was partially suppressed by treatment with Z-VAD-FMK, a pan caspase inhibitor. Therefore, Lico-E-induced oral cancer (OC) cell-specific apoptosis is mediated by the death receptor-dependent extrinsic and mitochondrial-dependent intrinsic apoptotic signaling pathways. In conclusion, these data suggested that Lico-E exhibits potential chemopreventive effects and warrants further developed as a chemotherapeutic agent against OC.

ResumoOs avanços no tratamento das neoplasias têm favorecido maiores taxas de sobrevida, aumentando a demanda da reabilitação dos pacientes com câncer, sendo a fisioterapia uma das áreas em amplo crescimento. Porém, essa população possui perfil, demanda e realidade bem diferentes, sendo necessário o conhecimento de suas características para melhor elaborar estratégias de atuação e condutas dentro da área de fisioterapia em oncologia. O objetivo deste estudo foi identificar o perfil clínico dos pacientes atendidos em um ambulatório de fisioterapia em oncologia. Foram analisados 105 prontuários para a coleta dos dados sociodemográficos e clínicos de um ambulatório de fisioterapia em oncologia do município de São Paulo, de agosto de 2008 a julho de 2010, sendo 11 (10,5%) excluídos. Para a análise estatística foi utilizado o teste de igualdade de duas proporções para avaliar a distribuição relativa das variáveis e Mann-Whitney para comparação destas. Dos 94 prontuários analisados, 63,8% (n = 60) eram do gênero feminino (p < 0,001), com faixa etária média de 56,3 ± 13,3 anos e IMC de 27,09 ± 5,88 kg/m 2 . O diagnóstico médico mais frequente foi o câncer de mama em 44,4% (n = 44; p < 0,05). A maioria estava entre o segundo e quinto ano (33%; n = 31) de evolução da doença. O tratamento cirúrgico foi o mais realizado (94,7%; n = 89; p < 0,001). O diagnóstico fisioterapêutico mais encontrado foi de alterações vasculares e linfáticas (87,2%; n = 82; p < 0,001). A queixa principal dos pacientes foi de dor (55,3%; n = 52) e edema (51,1%; n = 48; p = 0,559). Pode-se concluir que o perfil clínico dos pacientes se caracterizou por mulheres acima de 50 anos, com sobrepeso, em pós-operatório tardio de câncer de mama, apresentando dor e linfedema nos membros. Abstract: Cigarette smoking is clearly the most important head and neck cancer risk factor in this population. The reduced cancer risk due to physical activity was consistent with results from a pooled analysis of case-control studies.
