TNF-α Induces Apoptosis of Lactotropes from Female Rats

Abstract: TNF-alpha is involved in the regulation of normal tissue homeostasis affecting cell proliferation, differentiation, and death. We previously reported that TNF-alpha reduces anterior pituitary cell proliferation and PRL release in an estrogen-dependent manner. In the present project we studied the induction of apoptosis by TNF-alpha in anterior pituitary cells from female rats. TNF-alpha (50 ng/ml) decreased the viability of anterior pituitary cells. Incubation with TNF-alpha for 24 h increased the percentage o… Show more

“…TNF-␣-induced cellular shrinkage and atypical structure of nuclei in MCF7 cells as has been described previously for fibroblast (Luschen et al, 2000) and neuronal cells (Candolfi et al, 2002). It was difficult to observe the chromatin clumping (typical for apoptosis) in TNF-␣-treated cells due to shrinkage of the nuclei as opposed to this feature readily apparent in paclitaxel-treated cells (our unpublished data).…”

“…We have previously validated the coexpression of plasmid constructs in MCF7 cells (Foster, 2003). Morphological changes in the nuclei were analyzed and condensed/shrunk nuclei were counted as apoptotic as opposed to normal round nuclei (Candolfi et al, 2002).…”

“…Interestingly, Candolfi et al had reported that Fas activation induces apoptosis of anterior pituitary cells from female rats, suggesting that FasL is involved in the maintenance of tissue homeostasis in the anterior pituitary gland and could have potential benefits for the treatment of pituitary adenomas (18)(19)(20)(21). In the present study, we detected the expressions of Fas and DcR3 on 3 pituitary adenoma cell lines.…”

“…We have previously reported that TNFR1 and Fas activation induce apoptosis of anterior pituitary cells from female rats (Candolfi et al 2002, Jaita et al 2005b suggesting that TNF-and FasL are involved in the maintenance of tissue homeostasis in the anterior pituitary gland and could have potential benefits for the treatment of pituitary diseases. Although major advances have been made in the therapy of pituitary tumors, only partial success has been possible and novel therapies are needed.…”
