TNF-α gene polymorphisms in Iranian Azeri Turkish patients with Behcet’s Disease

Abstract: Genetic factors that predispose individuals to Behcet's disease (BD) are considered to play an important role in the development of the disease. The serum level of tumor necrosis factor (TNF) is elevated in patients with BD, and a dramatic response to anti-TNF-alpha antibody treatment further supports the role of TNF in BD. We investigated the distribution of TNF-alpha promoter -1031T/C and -308G/A polymorphisms in 53 BD patients of Iranian Azeri Turks and 79 matched healthy controls, via the PCR-RFLP techniqu… Show more

“…The frequencies of TNF-α -238 G/G and G/A genotypes were 6.2% and 93.8% in BD patients, 6.3% and 93.7% in healthy controls and the allele frequencies of TNF-α -238G were 0.53 in BD patients and 0.54 in healthy controls ( P = 0.500; OR = 0.96; 95% CI = 0.53–1.73). In previous study, we have already shown a significant association of BD with TNF-α -1031C allele ( P = 0.001; OR = 3.08; 95% CI = 1.73–5.47) in the same patients that we further studied in this research (21). …”

“…The lower frequency of allele A of TNF-α (-308G/A) has been reported in North European Caucasian and Korean patients with CD or UC as compared to healthy controls [103]. Although the frequency of allele G was reported to be slightly higher in Iranian Azeri Turkish IBD patients, it did not reach statistical significance [110]. Further, no association of TNF-α(-308G/A) polymorphism with IBD susceptibility was found in Australian [82], Brazilian [60,130], Canadian [100], Chinese [106,131], Czech [132], French [133], Indian [108], Korean [115], Newfoundland [27], Spanish [126], and Turkish [120] populations.…”

“…In large series, -308G/A and 238 GA polymorphisms of TNF-α were associated with asthma, systemic lupus erythematosus and juvenile RA in pediatric Mexican population [21]. Bonyadi [22] determined that allele and genotype distributions of -1031T/C polymorphism was significantly different in Behçet's disease of Iranian Azeri Turks. Vargas-Alarcon reported that -308A/G allel was increased in undifferentiated spondyloarthropathy group [23].…”
