Thiol–ene micropillar array electrospray ionization platform for zeptomole level bioanalysis

Abstract: A micropillar array electrospray ionization (μPESI) platform fabricated from thiol-enes with 56 individual polyethylene glycol coated μPESI chips for bioanalytical mass spectrometry is introduced. Bioanalysis capability is shown by measurement of a protein, a protein digest and a cell lysate sample. The thiol-ene polyethylene glycol (PEG) coated μPESI chip allows the use of a wide range of aqueous-organic solvent compositions and provides a detection limit at 60 zeptomole level (6 × 10 mol) for a peptide stand… Show more

“…18 To increase detection sensitivity (and circumvent any issues and challenges with optical detection), emitters for electrospray ionization mass spectrometry (ESI-MS) were developed using replica molding of TE. 21,99,122 Initially, the emitter taper was made in 2D, but for robust and long-term spray stabilities sharp emitter apexes are desired, and an improved version with a three-dimensional tapered geometry was developed (Figure 6C). 21 These 3D emitters lasted for more than a month before mechanical deterioration and showed good spray stabilities for at least 3 h, providing a relative standard deviation of 8% for the baseline over 15 min.…”

“…We speculate that this is due to reduced etching times and complete dissolution of the sacrificial layer, i.e., fewer residues. SU-8 devices released with the AZ 15nXT as sacrificial layer behave consistently compared to the devices which are released by other methods [18,19,31,39].…”

“…It provides a unified view of large-scale resources on the Internet in encyclopedia mode to realize an integrated search model of multiplatform online resources across the network. The literature [23] argues that the regional education big data platform should have context awareness, the full recording of natural interaction data, semantic aggregation of learning resources, and automatic content transcoding and format adaptation. The core modules such as subject knowledge base, cognitive model and inference engine established in the platform are used to analyze the interaction data of the learning process and personalize the learning resources and learning services.…”
