Thidiazuron: an efficient plant growth regulator for enhancing Agrobacterium-mediated transformation in Petunia hybrida

Abstract: Efficient shoot regeneration and Agrobacterium-mediated genetic transformation systems were developed for Petunia hybrida cv. Mitchell. Leaf explants of petunia were cultured on Murashige and Skoog (MS) medium with different concentrations of thidiazuron (TDZ) without auxin. The highest frequency of shoot regeneration (52.1%) and mean number of shoots per explant (4.1) were obtained on medium containing 2 mg l -1 TDZ. Leaf explants inoculated with Agrobacterium tumefaciens strain EHA101/pIG121Hm harboring ß-gl… Show more

“…Thidiazuron (TDZ) has been shown to enhance the frequency of shoot organogenesis in cotyledon explants (Vogel and Macedo 2011;Thirukkumaran et al 2009), which is in agreement with our findings as well. For P. pubescens, maintaining shoot regenerants on SEI medium containing TDZ for periods longer than 3-5 weeks resulted in the recovery of short and compact shoots (\1 cm in length, Fig.…”

“…It is used to indicate transient and stable transformations (Serres et al 1992), and to estimate the relative contribution and strength of promoter elements in the transcriptional activation. Most recently, GUS histochemical assay was utilized in the production of markerfree Petunia hybrida and tomato (Khan et al , 2011, and in the production of a more efficient Agrobacterium tumefaciens-mediated transformation protocol in melon (Ntui et al 2010), in Lilium (Azadi et al 2010), in Petunia hybrida (Thirukkumaran et al 2009), in pineapple (Gangopadhyay et al 2009), in Piper colubrinum (Mani and Manjula 2011), in sesame (Yadav et al 2010), in…”

“…Previous studies reported a positive effect of Kin on regeneration of many other plant species such as Salix viminalis (Grendysz et al, 2017), and cumin (Tawfik and Noga, 2002). Many other studies have reported successful shoot regeneration from petunia leaf explants when cultured on medium containing TDZ alone (Abu-Qaoud, 2012, Thirukkumaran et al, 2009. They attributed that to the effect of TDZ when included in the medium resulting in a balanced ratio of endogenous PGR that allows for specific modes of regeneration to take place, which is likely dependent on concentration as well as the plant species.…”
