Thermal hazard investigation of a pharmaceutical intermediate

Abstract: During the process development of a pharmaceutical advanced intermediate, new chemistry using an aqueous workup resulted in the discovery of a new hydrated crystal form of this intermediate. This hydrated crystal form showed thermal instability at a relatively low temperature compared to the original anhydrous form. Thermal stability and associated process safety hazards were investigated in order to ensure process safety and transportation safety of this intermediate.

“…The mines in the central and eastern mining areas have introduced deep mining before, and the current mining depth of deep mines is still expanding [1,2]. However, due to the expansion of mining depth, the temperature of deep mine shafts continues to rise, which seriously affects normal safe production [3,4]. Thermal damage in deep mine development is an objective reality, and creating a cool working environment is also an issue that needs to be improved in China's domestic mine development [5,6].…”

“…Hydroxyurea Intermediate. As fully described in a previously published paper, 17 the original process to synthesize and isolate a hydroxyurea intermediate used dry organic solvents, yielding a crystalline anhydrate form. This anhydrate form showed an exotherm initiation temperature of 120 °C from ARC testing (Figure 6) by EPSE, which does not present significant operational hazards for the scale-up.…”
