This study evaluated the in vitro activity of the arylaminoartemisinin GC012, readily obtained from dihydroartemisinin (DHA), against clinical strains of Helicobacter pylori (H. pylori) with different antibiotic susceptibilities in the planktonic and sessile state. The activity was assessed in terms of bacteriostatic and bactericidal potential. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined by the broth microdilution method. After treatment with GC012, all bacterial strains showed significantly lower MIC and MBC values compared to those of DHA. The effect of combination of GC012 with antibiotics was examined using the checkerboard method. GC012 displayed synergistic interactions with metronidazole, clarithromycin, and amoxicillin in all the strains. The antibiofilm activity was evaluated via crystal violet staining, AlamarBlue® assay, colony-forming unit count, and fluorescence microscopy. At ½ MIC and ¼ MIC concentration, both GC012 and DHA inhibited biofilm formation, but only GC012 showed a minimal biofilm eradication concentration (MBEC) on mature biofilm. Furthermore, both compounds induced structural changes in the bacterial membrane, as observed by transmission electron microscopy (TEM) and scanning electron microscopy (SEM). It is thereby demonstrated that GC012 has the potential to be efficacious against H. pylori infection.

Most of the plants are essential due to their medicinal properties and have contributed to the origin and evolution of many traditional herbal therapies. Rheum is also one of the famous and known medicinal plants of Kashmir Himalaya. It ranges from an altitude of 1700-5500 m. Its Roots are long and stout. The stem is erect, hollow, sulcate, glabrous, or strigose. It is native to Asia-Temperate to Asia-Tropical, from China to India, Nepal, and Pakistan. In India, its found in Himachal Pradesh, Jammu and Kashmir, and Uttar Pradesh. Rhubarb is the English name of the genus Rheum (family Polygonaceae). Emodi, Webbianum and Spiciforme are three species of Rheum found in Kashmir Himalaya. Rheum species are wellknown medicinal plant and is found on high altitudes. Its leaves and stalks are taken as food and cooked as vegetables. It's used as medicine and is generally proven more effective against certain ailments like antibacterial, anticancerous, anti-inflammatory, rheumatoid diseases, stomachache, and intestinal problems. It is also used for the treatment of indigestion, abdominal disorders, boils, wounds and flatulence. But Rheum has become endangered due to exploitation. Active compounds like aloe-emodin, rhein, emodin, chrysaphanol and physicion are found in this plant which is anti Cancerian agents. High molecular diversity is also observed in all the three species of Rheum.

Background Dental caries is a major oral health problem, which associates with cariogenic bacteria. Streptococcus mutans and Lactobacillus acidophilus are facultative anaerobic bacteria that are found in tooth decay. Accordingly, neem leaf extract was developed due to its great anti-microbial property against many bacteria. The aim of this study was to determine anti-cariogenic properties of neem leaf extract in a novel paste preparation. Material and Methods The neem extract was derived from maceration of dry neem leaves in ethanol for 48 h. The ethanolic extract was subjected to chemical identification using GC-MS. Neem pastes were prepared from ethanolic extract mixed with polyethylene glycol paste with or without zinc oxide. S. mutans and L. acidophilus test were initiated at bacterial concentration of 108 CFU/ml. The antibacterial activity was then performed by disc diffusion method following by minimum bactericidal concentration (MBC) technique. Results GC-MS result displayed 35 compounds. Compounds found in the extract were n-Hexadecanoic acid (31.18%), Hentriacontane (18%), Phytol (16.79%). Disc diffusion showed that ethanolic extract and neem pastes inhibited growth of both bacteria. For MBC, neem paste with zinc oxide at concentration of neem 0.4 mg/ml was the most effective concentration on inhibiting S. mutans growth. Neem pastes and ethanolic extract at concentration of neem 6.25 mg/ml inhibited L. acidophilus growth. Conclusions The ethanolic neem leaf extract and novel neem pastes had antimicrobial effect on both S. mutans and L. acidophilus . By this property, neem paste could be developed for the application in dental field, i.e. pulp capping. Key words: Neem, Azadirachta indica, antimicrobial, cariogenic bacteria.
