Theoretical and practical importance of plasma levels of haloperidol. correlations with clinical and computerized EEG data

Abstract: 1. Haloperidol plasma levels, after long acting haloperidol i.m. injection, are analyzed and compared with clinical évolution and EEG modifications. 2. ';ialoperidol plasma levels were determlned by radioiïïim'onoassay. Clinical status vas assessed weekly and computerized EEG were performed daily from day 3 to 7 after halo peridol decanoate injection. 3. The olasraa leveldose relationshlp during oral treatment and 4 weeks after haloperidol decanoate injection are very similar. After injection, the olasma level… Show more

“…For haloperidol decanoate, there appears to be a relatively high correlation between dose and plasma level (De Buck et al 1981;Reyntjens et al 1982;De Cuyper et al 1986). However, the relationship of plasma level to dose differs among the LINS.…”

“…It seems that SO far not many pharmacokinetic studies with human plasma monitoring have been carried out. In two studies a positive correlation between plasma haloperidol level and effect was found (236,237), whereas in other studies a positive correlation was lacking (231,238). The literature does not seem to include any studies in which the relation between maximum and minimum concentration in the dosage interval has been directly calculated.…”

“…Galderisi [6] reported that responders tended to exhibit greater slowing than nonresponders. In addition, EEG slowing in responders was found to be related positively to plasma haloperidol level [19,21] . In addition to treatment with conventional neuroleptics, Knott et al [7] reported that increments in relative theta power at the end of 6 weeks of clozapine treatment were paralleled by clinical improvement as demonstrated by PANSS symptom dimensions, supporting our observation that the increments in theta activity during treatment pointed to a good outcome.…”
