Theophylline: a review of population pharmacokinetic analyses

Abstract: This review concludes that body weight and age were the most important factors associated with the clearance of theophylline in paediatric patients. Body weight, age and smoking were most frequently used to estimate the clearance of theophylline in adults. Future studies are warranted to detect the influence of new factors, such as cytochrome P450 (CYP) 1A2 gene polymorphisms, on the pharmacokinetics of theophylline because some pharmacokinetic variability was not fully explained.

“…Clearly, this means that the results of our meta-analysis do not allow the placement of xanthines within the global COPD therapeutic framework. However, theophylline, being cheap and widely available, remains one of the most widely prescribed drugs for COPD treatment in developing countries [60]. Doxofylline, which presently is used in some regions of the world, can offer an alternative safer treatment, because of the lower risk of adverse events when compared with conventional "high-dose" theophylline, making blood monitoring unnecessary.…”

“…A. niger, Aspergillus tubingensis, Aspergillus fumigatus, Aspergillus acidus, Aspergillus awamori, A. tamarii, Blastobotrys adeninivorans, Candida tropicalis, Fusarium graminearum, Pichia farinosa, Rasamsonia byssochlamydoides, Rasamsonia emersonii, Rasamsonia cylindrospora, Rhizomucor pusillus, Rhizomucor tauricus and Thermomyces lanuginosus have been detected in pu-erh tea [28][29][30][31]. Theophylline has several applications in therapeutics, especially as anti-asthmatic, anticancer, anti-cellulite and combinatorial drug [32][33][34]. Caffeine content fluctuates during the SSF, which has associated with the fungi appearing in SSF [35][36][37][38][39].…”

“…However, in rising countries, Theophylline (THP) is at a halt the first-line treatment in patients with asthma and COPD, because it is low-priced and widely accessible. A growing amount of confirmation has recommended that low-dose THP has anti-inflammatory and immune modulatory effects in asthma and COPD and thus, THP has fascinated a large amount of awareness and importance [12,13]. THP fast metabolizers, as are started especially in the middle of children and smoking adults, may necessitate a further, regular interval than once-a-day dosing, and greater fluctuations in theophylline levels should be predictable [14].…”
