The β-glucan receptor dectin-1 functions together with TLR2 to mediate macrophage activation by mycobacteria

Abstract: Pattern recognition receptors (PRRs) play an essential role in a macrophage's response to mycobacterial infections. However, how these receptors work in concert to promote this macrophage response remains unclear. In this study, we used bone marrow-derived macrophages isolated from mannose receptor (MR), complement receptor 3 (CR3), MyD88, Toll-like receptor 4 (TLR4), and TLR2 knockout mice to examine the significance of these receptors in mediating a macrophage's response to a mycobacterial infection. We dete… Show more

“…Levels of mRNA were similar in MF from R17 and DA for TLR4 (a receptor for LPS) and for TLR2 (a -glucan receptor). 21 In contrast, mRNA levels were higher in R17 MF for Dectin-1 (a b-glucan receptor) and for CD163 (a scavenger receptor for hemoglobin-haptoglobin complexes). 22 Thus, APLEC indeed influences MF mRNA levels of other innate immunity receptors.…”

“…Dectin-1, a member of the C-type lectin receptor family, is expressed on the cell surface of phagocytes, acting as a sensor of glycans in fungi (e.g., Candida albicans) or mycobacteria (Hardison and Brown 2012;Rothfuchs et al 2007;Yadav and Schorey 2006). Activated dectin-1 induces the expression of various genes through NF-jB, leading to the production of cytokines that initiate the T helper 1 (Th1) or Th17 responses (Carvalho et al 2012;Gow et al 2007).…”

“…Dectin-1 is considered the main receptor for β -glucans (Brown et al 2002), and in cooperation with tool-like receptor 2 (TLR2) plays a role both in the activation of macrophages in response fungal and mycobacterial infections and participates in absorption of β -glucans from the gastrointestinal tract (Yadav and Schorey 2006;Suram 2006;Reid et al 2009). …”
