The vascular endothelial growth factor (VEGF) +405 G/C polymorphism and its relationship with recurrent implantation failure in women in an IVF programme with ICSI

Abstract: Purpose Successful embryo implantation depends on trophoblast proliferation, migration and, lastly, invasion of the endometrium (to anchor the trophoblast to the uterus). This invasion is mediated by locally produced soluble factors. Of these, vascular endothelial growth factor (VEGF) is the best characterized regulator of angiogenesis. Here, we investigate the association between the VEGF+405 C/G genotype and the recurrence of embryo implantation failure in women undergoing in vitro fertilization (IVF) progra… Show more

“…There have been several previous reports that VEGF polymorphisms are associated with the development and prognosis of a variety of obstetrical and gynecological diseases (17)(18)(19). Additionally, there have been reports that the VEGF -1154G>A and -634C>G polymorphisms are associated with the occurrence of RIF (20)(21)(22)(23), however, only a small number of studies have evaluated the association between other functional VEGF polymorphisms and the incidence rate of RIF (24,25).…”

“…This might be necessary for proper interaction between the endometrium and the conceptus prior to implantation in bovine. In humans, it is known that the conceptus-derived VEGFA induces vasodilatation and angiogenesis [27]. Additionally, the expression of VEGFA and VEGFR-1 in the human conceptuses further supports the likely regulatory role of VEGFA on conceptus proliferation, invasion and associated metabolic activities during implantation [27].…”

“…Furthermore, microRNA-374 was reported to activate the Wnt/β-catenin pathway, which is associated with implantation [28]. The four miRNAs that we investigated are known to bind the 3’UTRs of fibrinogen, VEGF, and KDR [13], all of which affect implantation [11].…”

“…VEGF functions to increase cell proliferation, migration, and differentiation and is highly polymorphic [10]. VEGF has been associated with RIF after IVF-ET with intracytoplasmic sperm injection [11]. Abnormal maternal expression of VEGF may be associated with abnormal angiogenesis during implantation [12].…”
