The unexpected structures of hepatitis C virus envelope proteins

Abstract: Hepatitis C virus (HCV) envelope proteins are essential not only for maintaining the viral life cycle, but also for evading the host's immune response and in clinical intervention. A thorough understanding of HCV envelope proteins depends on the availability of detailed structural information. Two crystal structures of the E2 core portion and of the E2 ectodomain, and one structure of the N-terminus of E1 ectodomain have shed new light on the complexity of HCV envelope proteins. In addition, the full-length E1… Show more

“…The core of E2 has been solved in complex with antibodies [2830], where E2 adopts a central immunoglobin-like fold formed by β-sheets surrounded by short α-helices dispersed in loops. However, both E1 and E2 used for structural studies are truncated proteins, part of the ectodomains are missing in the solved structures [31, 32]. Meanwhile, several groups have been trying to co-express E1E2 heterodimer to ensure the correct folding of the intact proteins as the folding and maturation of E1 and E2 may depend on each other [3336].…”

“…HCV is a single stranded RNA virus whose genome has single open reading frame (ORF) 9600 bp long ( Table 1 ) [ 37 ]. The N -terminal part of the ORF encodes structural proteins including core and envelope glycoproteins, whereas the remaining part of the ORF encodes for the nonstructural proteins [ 40 ].…”

“…It is a viral infection characterized by the inflammation of liver parenchyma [7]. Hepatitis C Virus is a single-stranded enveloped RNA virus belongs to the genus Hepacivirus of Flaviviruses [8]. Seven different genotypes (1-7) have been isolated which are identifiable on the basis of their nucleotide sequence [9].…”
