The type three secretion system effector protein IpgB1 promotes Shigella flexneri cell-to-cell spread through double-membrane vacuole escape

Abstract: S. flexneri is an important human pathogen that causes bacillary dysentery. During infection, S. flexneri invades colonic epithelial cells, hijacks the host cell cytoskeleton to move in the cytosol of infected cells, and spreads from cell to cell through formation of membrane protrusions that project into adjacent cells and resolve into double membrane vacuoles (DMVs). S. flexneri cell-to-cell spread requires the integrity of the bacterial type three secretion system (T3SS). However, the exact role of the T3SS… Show more

“…Enterobacteriaceae are widely distributed in nature; have a broad host range; and are parasitic or symbiotic in humans, animals, and plants. Some genera in the family Enterobacteriaceae, such as Escherichia ( 51 ), Salmonella ( 52 ), Shigella ( 53 ), and Klebsiella ( 54 ), belong to the common opportunistic pathogens. Ruminococcaceae are the main microorganisms that convert primary bile acids into secondary bile acids and are also involved in polysaccharide degradation ( 55 ).…”

“…Images were deconvolved using a Richardson-Lucy algorithm with 20 iterations and all slices within a stack were collapsed into a single image based upon the maximum intensity of pixels in each slice using NIS elements software. Protrusion duration was measured by quantifying the amount of time the bacteria were in each protrusion stage (54).…”

“…When the cytosolic niche of Shigella is eventually compromised, cell-to-cell spread is facilitated by actin polymerization by the autotransporter IcsA [ 211 , 212 ]. Double membrane vacuoles (DMVs) containing bacteria during intercellular spread can be degraded by IpgB1 [ 213 ]. As S. sonnei exhibits reduced invasion into epithelial cells [ 187 , 214 ] it will be interesting to evaluate whether it is also adapted to intracellular survival.…”
