The aim of this study was to evaluate the probiotic potential and the viability in fermented oat dairy beverage of Lactobacillus curvatus isolate P99. Probiotic potential was assessed through the survival in simulated gastrointestinal tract conditions, autoaggregation, coaggregation, and hydrophobicity capacity and antimicrobial activity. The safety aspects of the isolate were evaluated for application in food, and these were determined by phenotypic and genotypic tests. Isolate P99 was able to survive in simulated gastrointestinal tract conditions and showed hydrophobicity, autoaggregation, and coaggregation with Listeria monocytogenes. Moreover, the isolate and its cell free supernatant showed antimicrobial activity against Gram‐positive and Gram‐negative microorganisms. L. curvatus P99 showed in vitro probiotic potential, fulfilled safety aspects, and remained viable for 35 days in fermented oat dairy beverage stored at 4°C. Moreover, probiotic fermented oat dairy beverage demonstrated high acceptability. Therefore, isolate P99 can be considered a promising microorganism for the manufacture of probiotic foods. Practical applications Probiotics are microorganisms capable of providing health benefits to the host. However, isolates with probiotic potential from foods, when used as adjuvant cultures in foods, promote desirable sensory attributes and ensure microbiologically safe food. In this study, we found that Lactobacillus curvatus P99 presented probiotic potential and remains viable for 35 days when used in the production of fermented oat dairy beverage. Therefore, isolate P99 can be considered a promising microorganism for the manufacture of probiotic foods.

Aims/Introduction: Gut microbiota have various effects on human health. Some previous reports have shown that gut microbiota change during pregnancy and affect metabolism, but others have shown that microbiota do not change. Here, we examined the gut microbiota and glycoalbumin levels of 45 healthy Japanese women during pregnancy. Materials and Methods: We carried out 16S rRNA gene sequencing analyses of maternal stool samples and compared the gut microbiota composition of samples from women in early and late pregnancy. We also examined the association between gut microbiota and maternal characteristics, including glycoalbumin. Results: Microbiota composition in early and late pregnancy did not differ, according to principal coordinate analysis of weighted and unweighted UniFrac distances. Shannon indices were not different between early and late pregnancy. The proportion of one phylum, TM7, significantly decreased in late pregnancy compared with early pregnancy, but the proportions of other major phyla did not change. The Shannon index of late pregnancy was negatively associated with pregestational body mass index and positively correlated with glycoalbumin level, with adjustment of covariates. Conclusions: We concluded that Japanese women did not show obvious differences in gut microbiota during pregnancy, except for TM7, and that the diversity of gut microbiota might affect maternal metabolism. As this study had limited statistical power, further large-scale studies are required.

Obesity and its related complications have become increasingly serious social problems that threaten people's lives. Lactic acid bacteria (LAB) have shown an effective influence on preventing obesity in animal experiments and human clinical trials, but its exact mechanism remains unclear. In this review, LAB and several of its metabolites are taken as the point of penetration. It is aimed to summarize the relationship between obesity and gut microbiota, the efficacy of LAB, and its potential mechanisms of preventing obesity and lipid metabolism. LAB intervention can regulate gut microbiota and improve intestinal permeability which is damaged by a high-fat diet or lipopolysaccharide. Besides this, some metabolites produced by LAB can be involved in regulating host lipid metabolism-related pathways. Short-chain fatty acids, linoleic acid, and gammaaminobutyric acid, taken as representatives here, can participate in the regulation through AMP-activated protein kinase or peroxisome proliferator-activated receptor pathway and bowel-brain axis. Therefore, the ingestion of LAB may become a potential adjunctive treatment for obesity in future.
