The TIE1 Transcriptional Repressor Links TCP Transcription Factors with TOPLESS/TOPLESS-RELATED Corepressors and Modulates Leaf Development inArabidopsis

Abstract: Leaf size and shape are mainly determined by coordinated cell division and differentiation in lamina. The CINCINNATA (CIN)-like TEOSINTE BRANCHED1/CYCLOIDEA/PCF (TCP) transcription factors are key regulators of leaf development. However, the mechanisms that control TCP activities during leaf development are largely unknown. We identified the TCP Interactor containing EAR motif protein1 (TIE1), a novel transcriptional repressor, as a major modulator of TCP activities during leaf development. Overexpression of T… Show more

“…In this complex, Cyc8 functions as a Tup1 adaptor (Tzamarias and Struhl, 1994). In plants, many transcription factors can bind TPL directly (Causier et al, 2012), but others, as in Cyc8-Tup1, recruit TPL via molecular bridges such as NINJA, JAZ, Aux/IAA, and TIE1, which links TEOSINTE BRANCHED1/CYCLOIDEA/PCF (TCP) transcription factors to TPL (Tao et al, 2013), including the KIX proteins presented here.…”

“…To verify whether NNC1 is a transcriptional repressor, we performed a classical GAL4/UAS-based system assay to study the transcriptional repression of a protein (Tao et al, 2013). As shown in Figure 6C, the GAL4 DNA binding domain (G4DBD) that binds to the six copies of GAL4 UAS can activate the GUS gene expression.…”

“…The histochemical GUS staining analysis was described previously (Tao et al, 2013). Briefly, tissues of pEXB1-GUS transgenic lines or DR5-GUS plants were immerged in 90% acetone for 30 min on ice and washed by phosphate buffer twice.…”
