The WRKY Transcription Factor WRKY71/EXB1 Controls Shoot Branching by Transcriptionally Regulating RAX Genes in Arabidopsis

Abstract: Plant shoot branching is pivotal for developmental plasticity and crop yield. The formation of branch meristems is regulated by several key transcription factors including REGULATOR OF AXILLARY MERISTEMS1 (RAX1), RAX2, and RAX3. However, the regulatory network of shoot branching is still largely unknown. Here, we report the identification of EXCESSIVE BRANCHES1 (EXB1), which affects axillary meristem (AM) initiation and bud activity. Overexpression of EXB1 in the gain-offunction mutant exb1-D leads to severe b… Show more

“…Mesophyll protoplast preparation and transfection were performed according to a previously reported method (Yoo et al, 2007). For transient expression assays in tobacco leaves, leaves of Nicotiana benthamiana were transiently transformed and examined as previously described (Guo et al, 2015). Biological replicates represent the results of three independent assays.…”

“…WRKYs are TFs that have been reported to induce dwarfing (Cai et al, 2014;Guo et al, 2015;Li et al, 2016). We screened five dwarfing rootstocks and five nondwarfing rootstocks of apple and detected the expression of 29 WRKY genes that may function in plant development.…”

“…In transgenic rice, inhibition of OsWRKY78 resulted in a semi-dwarf plant displaying a small-kernel phenotype due to reduced cell length . OsWRKY11 overexpression reduced the height of transgenic rice plants, leading to plants with a dwarf stature (Cai et al, 2014;Guo et al, 2015). The plant WRKY TFs are characterized by the presence of one or two WRKY domains, including the highly conserved WRKYGQK sequence, and a zinc-binding motif (Duan et al, 2007;Zhang et al, 2011).…”

“…RAX proteins have additional roles during reproductive development (Keller et al 2006;Müller et al 2006). Recently, gain-of-function of five closely related WRKY transcription factors each promoted axillary meristem initiation; one of these, EX-CESSIVE BRANCHES1 (EXB1/WRKY71), positively regulates RAX1, 2, and 3 transcription (Guo et al 2015).…”
