The structure of the yeast Ctf3 complex

Abstract: Kinetochores are the chromosomal attachment points for spindle microtubules. They are also signaling hubs that control major cell cycle transitions and coordinate chromosome folding. Most well-studied eukaryotes rely on a conserved set of factors, which are divided among two loosely-defined groups, for these functions. Outer kinetochore proteins contact microtubules or regulate this contact directly. Inner kinetochore proteins designate the kinetochore assembly site by recognizing a specialized nucleosome cont… Show more

“…Major recent progress has been made in understanding the structure of the budding yeast CCAN (also referred to as Ctf19 complex) ( figure 4 b ) [ 144 146 ]. These structures contain CENP-I Ctf3 , CENP-H Mcm16 , CENP-K Mcm22 , CENP-L Iml3 , CENP-N Chl4 , CENP-P Ctf19 , CENP-O Mcm21 , CENP-Q Okp1 , CENP-U Ame1 , CENP-T Cnn1 and CENP-W Wip1 , as well as Nkp1 and Nkp2, which apparently lack human homologues.…”

“…As a sole HEAT protein and an essential subunit in kinetochore, the structure and function research of CENP‐I would be useful to understand kinetochore assembly and chromosome segregation. The recent Cryo‐EM structures had revealed the multiple interactions of CENP‐I and other subunits . However, the intramolecular interaction and structure details of CENP‐I itself have not been analyzed yet.…”

“…We previously reported the N‐terminal half structure of fungal CENP‐I that forms a ternary complex with C‐terminal heterodimer of fungal CENP‐H/K . The recent cryo‐EM structures revealed another half of Ctf3 (CENP‐I homolog), which forms complex with N‐terminal heterodimer of Mcm16/22 (CENP‐H/K homolog) . However, the intramolecular interaction and structure details of CENP‐I are less discussed.…”
