The smooth muscle cell. III. Elastin synthesis in arterial smooth muscle cell culture.

Abstract: Primate arterial smooth muscle cells and skin fibroblasts were examined for their ability to synthesize elastin in culture. In the presence of the lathyrogen/~-aminopropionitrile, the smooth muscle cells incorporate [SH]lysine into a lysyl oxidase substrate that was present in the medium and associated with the cell layer. A component having a mol wt of 72,000 and an electrophoretic mobility similar to that of authentic tropoelastin was isolated from the labeled smooth muscle cells by coacervation and fraction… Show more

“…In general, elastin is synthesized by fibroblasts, but also by smooth muscle cells, endothelial cells, and cartilage cells [21][22][23][24]. Thus, fibroblasts may be one source of excessive elastin production in the tumor stroma [3,7].…”

“…These residues offer positive charges for binding with negative charges of glycosaminoglycan. In the absence of lysyl oxidase such electrostatic interaction could be important and prevent newly synthesized tropoelastin molecules from spontaneous random aggregation far from the cell surface [37,67]. Long and Tranquillo [2] studied the effect of TGF-h1 and insulin on elastogenesis in rat aortic smooth muscle cells.…”

“…Interestingly when smooth muscle cells are cultured in conventional tissue culture dishes, they are known to secrete both soluble (precursor) and insoluble elastin [37]. In addition, it has been reported that static stretching of vascular smooth muscle cells in culture dishes stimulated elastin biosynthesis as well as crosslinking [38].…”

“…Elastin is widely distributed in arteries, alveoli, skin, nuchal ligaments, and elastic cartilages. It has been confirmed that elastin is synthesized by smooth muscle cells in arterial walls [ 1 ], fibro blasts in lung alveoli [2], skin [1], nuchal liga ments [3], and by chondrocytes in elastic car tilages [4].…”
