Abstract. Sari YP, Kustiawan W, Sukartiningsih, Ruchaemi A. 2017. Short Communication: The potential of secondary metabolites of Myrmecodia tuberosa from different host trees. Nusantara Bioscience 9: 170-174. Ant-plants (Myrmecodia tuberosa Jack.) is a medicinal plant that could potentially inhibit cancer cell growth. Ant-plants is epiphytic plants whose commonly life was attached to the host tree. Several information from local people stated that ant-plants attaching to different host trees possesses different active compounds. The purpose of this study was to determine the secondary metabolites of each parts of ant-plants including leaves, stems and tubers from different tree hosts i.e mango and durian. Result from phytochemical analysis showed that ant-plants living in mango and durian trees positively contained the metabolic compounds including phenolics, flavonoids, alkaloids, saponins and steroid/triterpenoid. The Total of Phenolic Content (TPC) and the Total of Flavonoids Content (TFC) on the leaves of ant-plants was higher than that in tubers or stems of ant-plants derived from both host trees i.e mango and durian. The value of TPC and TFC of the ant-plants leaves derived from the durian host tree (319.33 ± 0.06 mg GAE/g and 272.33 ± 0.02 mg CE/g) was higher than those from mango host trees (172.80 ± 0.02 mg GAE/g and 162,83 ± 0.01 mg CE/g). Ant-plants, whose life was attached to a different host tree, had the same content of secondary metabolites although those quantities were different in each host trees. Therefore, ant-plants from different host trees could be used as a medicinal plant by concerning the amount of used extract for effectiveness medicinal purposes.

This current study is to evaluate and compare the antibacterial, antioxidant and anticancer activity of H. formicarum tubers from Setiu Wetland, Terengganu, Malaysia and inland forest of Muara Rupit, SumateraSelatan, Indonesia. The tubers were extracted using methanol and then sequentially fractioned using solvent partitioning by n-hexane, dichloromethane, and ethyl acetate. The phytochemical study showed that samples from both places possessed same phytochemicals content that was rich with phenols, tannins, steroids, and glycosides. Extract from Setiu exhibit antibacterial activity against all tested bacteria, while methanol extract from Muara Rupit was only active against Gram positive bacteria. All samples showed high DPPH free radical scavenging activity (IC50 less than 10 µg/ml). Via MTS assay, no cytotoxic activity of all samples was observed against HeLa cells. Only a fraction from Setiu Wetland showed very strong cytotoxic activity against MCF-7 cells (IC50 = 2 µg/ml) and its morphological features stained by Annexin-V/PI and DAPI showed that the cell death was mediated by apoptosis. Results revealed that H. formicarum tuber from Setiu wetland and Muara Rupit showed very good potency as antibacterial and antioxidant agents. The only sample from Setiu wetland has a good potency as anticancer against MCF-7 cells but not against HeLa cells.

Objective: Despite advanced treatment options available for colorectal cancer, many reported resistance and unresponsiveness to conventional chemotherapeutic agents. Therefore, it is urgent to discover a novel drug for colon cancer. Sarang Semut (Myrmecodia pendans), an Indonesian native plant, has been studied extensively due to its anti-cancer profiles. This study aimed to evaluate the anti-tumour activity of Sarang Semut in colon cancer cells. Methods:We evaluated cytotoxic activity of methanol extract as well as n-hexane and ethyl acetate fraction towards colon cancer cell lines (Caco-2 and HCT-116 cells) utilizing 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) assay. The most potent fraction was evaluated further in inhibiting cell survival using MTT assay and cell proliferation using trypan blue exclusion assay as well as a clonogenic assay. Results:Our data showed that the n-hexane fraction of Sarang Semut induces more cell death than the methanol extract and ethyl acetate fraction. Therefore, we analyzed the n-hexane fraction further and found that the inhibitory concentration 50% (IC50 Conclusion:The n-hexane fraction of Sarang Semut demonstrates a high potential antitumor activity in colon cancer cell line.) of the n-hexane fraction was 24 and 30 parts per million (ppm) for Caco-2 and HCT-116 cells, respectively. Moreover, it inhibited cell growth as well as cell colony formation, in particular, shown by the plating efficiency (P<0.05) and colony area per seed (P<0.01) of the control group were different to the treatment group.
