The role of total and high-molecular-weight complex of adiponectin in vascular function in offspring whose parents both had type 2 diabetes

Abstract: Aims/hypothesis: Adiponectin is an adipokine with insulin-sensitising and anti-atherogenic properties. We studied the role played by total adiponectin and by the bioactive high-molecular-weight (HMW) oligomeric complexes of adiponectin in vascular function in offspring whose parents both had type 2 diabetes, a population at high risk of diabetes and atherosclerosis. Methods: Total and %HMW adiponectin, the cytokines C-reactive protein, interleukin-6 and plasminogen activator inhibitor-1 (PAI-1), as well as lip… Show more

“…As described above, we noted a very strong correlation between serum HMW and total adiponectin (r ϭ 0.969), in agreement with a recent study in offspring whose parents both had type 2 diabetes using velocity sedimentation with Western blotting (r ϭ 0.72 between total adiponectin and the HMW-to-total adiponectin ratio) (20). This suggests that total and HMW adiponectin may be regulated synchronously.…”

“…Of the 3 distinct multimeric isoforms in the circulation, HMW adiponectin (12-to 18-mers) is believed to be the most biologically active (5,12,13 ). The HMW adiponectin concentration and/or percentage of HMW adiponectin are reported to be a better index of insulin sensitivity than total adiponectin (14,21 ).…”

“…However, none of the mentioned gene variants has been clearly associated with changes in plasma adiponectin, and total adiponectin levels have been reported not to be associated with IMT. 22 We cannot exclude that the high-molecular weight isoform of the adipokine, which is believed to carry most of the biological value, 46 may relate to some aspect of vascular structure or function.…”
