Comparison of Serum High–Molecular Weight (HMW) Adiponectin With Total Adiponectin Concentrations in Type 2 Diabetic Patients With Coronary Artery Disease Using a Novel Enzyme-Linked Immunosorbent Assay to Detect HMW Adiponectin

Abstract: Adiponectin (Acrp30), an adipocyte-derived protein, exists in serum as a trimer, a hexamer, and a high-molecular weight (HMW) form, including 12-18 subunits. Because HMW adiponectin may be biologically active, we measured it in serum using a novel enzyme-linked immunosorbent assay (ELISA) confirmed by gel filtration chromatography that the ELISA detected mainly adiponectin with 12-18 subunits, and we compared HMW with total adiponectin concentration in patients with type 2 diabetes. We next investigated the re… Show more

“…Briefly, serum lipoproteins were separated on a column containing diethylaminoethyl-ligand nonporous polymer-based gel by elution with a step gradient of sodium perchlorate concentration, and detected by a post-column reaction with a reagent containing cholesterol esterase and cholesterol oxidase. Serum total adiponectin was measured by ELISA obtained commercially from Otsuka Pharmaceutical Co., Ltd. (Tokyo, Japan) 5) , and HMW adiponectin was also measured by ELISA using a monoclonal antibody against human HMW adiponectin (Fujirebio, Tokyo, Japan) 13) . Serum insulin, total cholesterol (TC), TG, fasting plasma glucose (FPG), and hemoglobin (Hb)A1c were measured conventionally.…”

“…For estimation of insulin sensitivity, a homeostasis model assessment of insulin resistance (HOMA-IR) was calculated. Plasma total adiponectin and high-molecular-weight (HMW) adiponectin concentrations were measured by sandwich ELISA (Otsuka Pharmaceuticals, Tokyo, Japan and Fujirebio, Tokyo, Japan respectively) as previously described (18).…”
