A B S T R A C TIschemic heart disease is a common cardiac health problem. Despite the significant advances in prevention and treatment of this disorder, its incidences and complications are very serious. So, the search for more antioxidants and anti-inflammatory agents with cardioprotective effects is an urgent task. We aimed to evaluate the effects of a heme oxygenase 1 (HO1) inducer, hemin (HEM), on isoprenaline (ISO)induced myocardial damage. Forty-five Wistar albino rats were used. Animals were treated with HEM (25 mg/kg/day) i.p. for 5 days and injected with ISO (150 mg/ kg/day) i.p. on 4th and 5th day of the experiment. Detection of the role of ATPsensitive potassium channel (K ATP ) was performed by administration of glibenclamide (GP) (5 mg/kg/day) orally 2 h before HEM. Moreover, the role of endothelial nitric oxide synthase (eNOS) was detected by coadministration of Nitro-x-L-arginine (L-NNA) (25 mg/kg/day) for 5 days. The ISO group showed increase in heart weight, cardiac enzymes, tumor necrosis factor alpha (TNFa), and malondialdehyde (MDA) with decrease in reduced glutathione (GSH), HO1, and total antioxidant capacity (TAC). In addition, there were increases in Bcl-2 associated X protein (Bax) and cleaved caspase-3, but decreases in B-cell lymphoma-2 (Bcl-2) and eNOS. Moreover, the histopathological examination of the ISO group showed degeneration of the cardiac muscle fibers and marked infiltration of the inflammatory cells. The biochemical and histopathological changes induced by ISO were markedly ameliorated in the HEM plus ISO group. This protective effect was diminished with coadministration of GP or L-NNA; thus, K ATP and eNOS might mediate HEM cardioprotection. I N T R O D U C T I O NCardiovascular diseases including myocardial infarction (MI) are among the most severe illnesses that could lead to death. MI occurs due to the imbalance between oxygen supply and oxygen demand followed by myocardial hypoxia [1]. This imbalance with the excessive release of reactive oxygen species (ROS) is a major contributing factor in the development of MI. Free radicals attack membrane lipids and mediate a chain of reactions of lipid peroxidation associated with disruption of the structure and permeability of cell membranes [2,3].Isoprenaline (ISO), a synthetic b-adrenergic agonist, causes severe oxidative stress and decreases oxygen supply in the myocardium leading to infarct-like necrosis in the heart muscles. The objective of this study was to evaluate the effects of zearalenone (ZEA) and estradiol benzoate (EB) on stress injury and uterine development in post‐weaning gilts. Thirty healthy post‐weaning female gilts (Duroc × Landrace × Large White) aged 28–32 days were randomly allocated to three treatments as follows: (a) basal diet (Control), (b) basal diet plus 1.0 mg/kg purified ZEA (ZEA) and (c) basal diet plus 0.75 ml (1.5 mg) EB per pig at 3‐days intervals by intramuscular injection (EB). The serum estradiol (E2), the final and the increased vulvar area, uterine index, thickness of the myometrium and endometrium, and protein expression of heat shock protein 70 (HSP70) in ZEA group were higher than those in the control group (p < .05), but lower than those in the EB group (p < .05). The serum luteinizing hormone in ZEA group was lower than that of the control group (p < .05), but higher than that in the EB group (p < .05). Higher serum follicle‐stimulating hormone and progesterone were observed in the ZEA and control groups than those in the EB group (p < .05). The serum glutathione peroxidase activity in the ZEA group was lower than that in the control and EB groups (p < .001), and the malondialdehyde in the ZEA group was higher than that in the control and EB groups (p < .001). Moreover, the relative mRNA and protein expression of growth hormone receptor (GHR) and relative mRNA expression of HSP70 in the ZEA and EB groups were higher than those in the control group (p < .05). In conclusion, both ZEA (1.0 mg/kg) and EB (1.5 mg at 3 days intervals by intramuscular injection) stimulated vulvar swelling and uterine hypertrophy by disordering serum hormones and up‐regulating GHR expression, and induced stress by different mechanisms in this study. Furthermore, the observed up‐regulating HSP70 expression challenged by ZEA or EB may be part of the mechanism to resist stress injury.

Ischemia reperfusion injury (IR) is an unavoidable problem that frequently occurs during resection of liver tumors or liver transplantation. Diacerein is a chondroprotective agent used clinically for treatment of osteoarthritis. It exhibits anti-inflammatory, anti-apoptotic and antioxidant properties. So, in the current study, we hypothesized that diacerein may attenuate the partial warm hepatic IR injury in rats. Also, the study aimed to elucidate the possible underlying mechanisms. Rats were randomly divided into: sham, IR, and two diacerein pretreated groups. Rats were treated with diacerein 5mg/kg/day and 10mg/kg/day orally for 14 days and then, partial hepatic ischemia was performed for 30 min followed by reperfusion for 6 h. Liver function markers were determined. Markers of oxidative stress, apoptosis and inflammation were measured. Hematoxylin and eosin stained sections of liver tissue were examined as well as immunohistochemical stained sections for detection of nuclear factor kappa B-p65 (NF-κB-p65) expression. IR insult deteriorated the liver function and evoked inflammation, apoptosis and oxidative stress as well as histopathological derangements. Pretreatment with diacerein 10mg/kg improved the deteriorated liver function and the histopathological changes as well as attenuating the oxidative stress, apoptosis and inflammatory processes. Diacerein attenuates partial IR induced liver damages through anti-inflammatory, anti-apoptotic actions and inhibition of oxidative stress.
