The Role of H2S Regulating NLRP3 Inflammasome in Diabetes

Abstract: Nucleotide-binding oligomeric domain (NOD)-like receptor protein 3 (NLRP3) is a recently discovered cytoplasmic multiprotein complex involved in inflammation. The NLRP3 inflammasome contains NLRP3, apoptosis-related specific protein (ASC) and precursor caspase-1. The NLRP3 inflammasome is involved in many diseases, including diabetes. H2S is a harmful gas with a rotten egg smell. Recently, it has been identified as the third gas signal molecule after nitric oxide and carbon monoxide. It has many biological fun… Show more

“…Highly water soluble H 2 S exists primarily in three different forms, namely: S 2− , HS − and H 2 S. The form of the sulfide is dependent on the pH of the aqueous medium [4,5]. Serious physiological problems, like Down syndrome [6], diabetes [7], liver cirrhosis [8], and Alzheimer's [9], can arise from excess sulfide exposure. In addition to the industrial sources of H 2 S, the illegal use of sulfide additives in food such as sulfites, rongalite, and sodium sulfide can cause harmful effects on human health [10].…”

“…In one study, H 2 S could protect HaCaT human keratinocytes from methylglyoxal (MGO) induced damage and behavioral disorders by inhibiting apoptosis, alleviating intracellular ROS content, increasing mitochondrial membrane potential, and ultimately promoting cell adhesion and migration [93] . Meanwhile, exogenous H 2 S inhibits NLRP3 inflammasome mediated inflammatory response by inhibiting high glucose induced ROS production in epithelial cells [94] .…”

“…Although endogenous factors that can activate inflammasomes (Figure 2) have been uncovered, additional research is required to comprehend how these signals are direct determinants of diabetes-related macrovascular dysfunction [23,64,65]. While endogenous signals that can activate inflammasome assembly have been identified, further research is needed to understand how they are directly linked to diabetes-related macrovascular injury.…”
