Background: Hypertension and pregnancy cause placental ischemia which will produce the toxic free radicals that cause the oxidative stress and increase the malondialdehyde (MDA) level. Oxidative stress also decreases the Vascular Endothelial Growth Factor (VEGF) level. Extra Virgin Olive Oil (EVOO) contains antioxidants that can break free radical chain reactions. This study aims to determine the effect of the EVOO administration on the MDA and VEGF levels on the hypertensive pregnant rats. Methods: A post-test only control group design study was conducted on 30 pregnant rats consisting of a negative control group (K-), a positive control group (K+) and 3 treatment groups as a hypertension model (Pl, P2, P3), NaC1-induced hypertension model 6 % on day 6 to day 12 of pregnancy. All treatment groups were given EVOO except K+, from day 13 to day 19. On day 20, all the rats were executed. The MDA examination used a spectrophotometer and the VEGF examination used ELISA. The data was tested by using the one way ANOVA test which was statistically significant if p < 0.05. Results: The results of this study showed that there was a significant decrease on the mean of the MDA level after the EVOO administration, especially in the P3 group, which was 1,532 mmol/l (p 0.000) and a significant increase on the mean of the VEGF level after the EVOO administration, especially in the P3 group, which was 68,892 ng/l. (p 0.000). It means that there is an effect of the EVOO administration on the MDA and VEGF levels of hypertensive pregnant rats. Conclusion: The conclusion of this study is that the EVOO administration can decrease the MDA level and increase the VEGF level on hypertensive pregnant rats. Oxidative stress induced by hydrogen peroxide (H2O2) can lead to cellular damage, contributing to degenerative diseases and aging. Black rice bran is a functional food known for its antioxidant properties, which are crucial in reducing the adverse effects of oxidative stress and maintaining redox balance. In this study, we aimed to investigate the protective effect of the extract of black rice bran (EBRB) 'Sembada Hitam' on Vero cells against H2O2 toxicity. To evaluate the protective effect, a co-culture method was employed, and cell viability was assessed using the MTT assay. Additionally, cell growth was examined through trypan blue staining. Vero cells were exposed to different concentrations of H2O2 and EBRB for a 24-hour period. The results demonstrated that EBRB at concentrations of 15.625, 250, and 500 μg/mL exhibited a protective effect on Vero cells exposed to H2O2 at concentrations of 100, 200, and 400 μM, respectively. Notably, when Vero cells were treated with EBRB at concentrations of 250 or 500 μg/mL for five days in conjunction with H2O2 exposure at concentrations of 200 or 400 μM for 24 hours, a significant decrease in cell viability was observed on day 3. Based on the collective findings, it can be concluded that EBRB has the potential to protect Vero cells against H2O2 -induced toxicity, but primarily during a short-term incubation period. Overall, this study highlights the protective properties of EBRB against H2O2-induced cellular damage and emphasizes the importance of further investigations to fully elucidate the underlying mechanisms and potential long-term effects of EBRB on cell viability.

Preeklamsi merupakan penyebab utama morbiditas dan mortalitas baik maternal maupun perinatal. Tujuan: Menganalisis pengaruh pemberian likopen terhadap kadar sFlt-1 secara in vitro pada sel trofoblas model preeklamsi. Metode: Penelitian ini dilakukan secara eksperimental di Laboratorium Genetika Molekuler dan Kultur Sel Fakultas Kedokteran Universitas Padjadjaran selama periode 2017-2018. Sampel serum diperoleh dari RS. Dr. Hasan Sadikin Bandung, sesuai kriteria inklusi dan eksklusi. Kriteria inklusi adalah pasien yang telah didiagnosis preeklamsi berdasarkan pemeriksaan tekanan darah dan protein urin, dan kriteria eksklusi adalag pasien yang tidak datang kontrol atau tidak bisa dihubungi. Penelitian ini terdiri dari dua tahap, pertama ditentukan nilai LD50 likopen terhadap sel trofoblas kemudian dijadikan dasar nilai konsentrasi yang akan digunakan, selanjutnya dilakukan pemeriksaan sFlt-1 menggunakan metode Elisa pada kultur sel trofoblas kondisi normal dan preeklamsi dengan analisa data menggunakan analisis varians (Anova) dan uji Duncan untuk mengetahui perbedaan antar variabel. Hasil: Nilai LD50 adalah 250 ug/ml dan serial konsentrasi yang digunakan mulai dari 1,953 sampai dengan 125 ug/ml. Didapatkan kadar sFlt-1 menurun secara signifikan (p0,05) dari 39,5445 menjadi 31,1342 pg/ml pada sel trofoblas yang diinduksi dengan serum preeklamsi setelah pemberian konsentrasi likopen sebesar 15,625-31,25 ug/ml yang nilainya mendekati keadaan pada sel trofoblas yang diinduksi serum kehamilan normal. Pemberian likopen dapat menurunkan kadar sFlt-1 pada sel trofoblas preeklamsi. Simpulan: Likopen dapat merubah kadar sFlt-1 dan perubahannya dipengaruhi oleh konsentrasi dan jenis serum.Kata kunci: likopen, preeklampsi, sFlt-1
