The RhoGAP Stard13 controls insulin secretion through F-actin remodeling

Abstract: ObjectiveActin cytoskeleton remodeling is necessary for glucose-stimulated insulin secretion in pancreatic β-cells. A mechanistic understanding of actin dynamics in the islet is paramount to a better comprehension of β-cell dysfunction in diabetes. Here, we investigate the Rho GTPase regulator Stard13 and its role in F-actin cytoskeleton organization and islet function in adult mice.MethodsWe used Lifeact-EGFP transgenic animals to visualize actin cytoskeleton organization and dynamics in vivo in the mouse isl… Show more

“…STARD13 encodes the protein StAR-related lipid transfer protein 13, a Rho GTPase-activating protein closely associated with hepatic cancer ( 80 ). Besides being linked with actin cytoskeletal organization, this protein also seems to influence other biological activities ( 81 ), such as insulin secretion ( 82 ), and induction of mitochondrial phosphatidylglycerolphosphate synthase activation in response to ceramides ( 83 ). This may be particularly relevant in the adipocyte’s metabolic deregulation, as ceramides play an important role in glucose uptake and metabolism ( 84 ).…”

“…Finally, the values were normalized by the number of cells. For the analysis of filamentous actin (F-actin), the fluorescence intensity of F-actin in images on split channels were measured in ImageJ and normalized by the number of cells 38 . We performed morphological analysis from at least three randomly selected images per group in three independent experiments (containing total number of cells ranging from 1,089 to 1,521 cells per group).…”

“…3 C). F-actin, the major component of the cytoskeleton microfilament [19] , was displayed to be tentacle-like and more polarity detected by phalloidin staining in JUP-knocked down NCI-87 cells ( Fig. 3 D), which accompanied with notably decreased epithelial marker E-cadherin and β-catenin and enhanced mesenchymal marker Vimentin and Fibronectin in the JUP-knocked down NCI-87 cells ( Fig.…”
