The results of diethylstilboestrol therapy for recurrent and metastatic carcinoma of the male breast

Abstract: A retrospective survey has been made of 58 patients with recurrent and advanced male breast cancer treated with oral diethylstilboestrol. Fifty-five patients were suitable for assessment. Fourteen patients had an objective response and 7 had a partial response giving a total response of 21/55 patients (38%). The median remission for the objective responders was 7 years. Three patients are alive and free of disease; one has now been in remission for 13 years. It is suggested that diethylstilboestrol is a useful… Show more

“…Several reports exist showing mild to moderate success with a variety of agents. Estrogen therapy in the form of diethylstilboestrol has been shown to have a 38%-40% total response rate when given to patients with advanced or recurrent male breast cancer [25,26]. Cyproterone acetate, a synthetic steroid with antiandrogenic and progestational activity has also shown success.…”

“…92,93 Given the often deleterious side effects of these ablative techniques, other means of hormonal manipulation have been tried with mild to moderate success reported. A variety of agents have been tested, such as estrogen therapy in the form of diethylstilbestrol, 95,96 cyproterone acetate (a synthetic steroid with anti androgenic and progestational activity), 97 medroxy progesterone acetate, 98 androgens, 94,95 and luteinizing-hormonereleasing hormone agonists, alone or in combination with anti androgens. 99,100 Because of the success of tamoxifen for treating both advanced MBC (responses ranged from 25-80%), 92,94,101 and as an adjuvant treatment (5-year diseasefree survival increased from 28% to 56%, and 5-year overall survival from 44% to 61%), 102 tamoxifen was incorporated as the mainstay of hormonal therapy in MBC.…”

“…Although in a retrospective re view of 55 patients treated with diethylstilbestrol (DES), an overall response rate of 38% was reported [11], estrogens have been generally considered of lim ited usefulness for MBC [12], and exacerbation of the disease has sometimes occurred with these agents [13]. Our limited experience is somewhat better, with re gression of osseous and visceral metastases for 20 and 33 months in 2 of 5 cases treated with estrogenic compounds.…”
