S IX protein fractions have been reported to be present in the blood serum of the cock (McCully et al, 1959; McKinley et al., 1953;Heim and Schectman, 1954). These fractions were separated by zone electrophoresis on paper using aqueous veronal or borate buffer. The six protein zones were designated in order of decreasing mobility as: albumin, alpha-1-globulin, alpha-2-globulin, alpha-3globulin, beta globulin and gamma globulin. In addition to these six fractions. Brandt et al. (1951) found a fraction (pre-albumin) moving ahead of albumin in the sera of laying hens. This pre-albumin was apparently absent in the sera of males (Brandt et al., 1951;Heim and Schectman, 1954). In contrast to these findings, Kristjansson et al. (1963) using vertical starch gel electrophoresis found two protein fractions migrating in front of albumin in the sera of males. A third pre-albumin was only present in the sera of hens in production.The present study was conducted using polyacrylamide gel columns and disc electrophoresis to quantitatively evaluate the blood sera proteins of the adult male chicken.EXPERIMENTAL PROCEDURE Blood samples (3 cc.) were taken on different days by needle from the brachial vein of each of 12 individual New Hampshire males. These males were approximately one and one-half years of age at the time of initial sampling. Summary Ionized Calcium in Blood of Domestic Fowl: Dependence on Age and Sex In the present study concentration of ionized calcium and the correlation between total and ionized calcium in blood of domestic fowl and its dependence on age and sex were investigated. Furthermore a possible influence of blood sampling procedure on the measured values was assessed. Leghorn chickens of either sex, aged 3, 9, 25, and 80 weeks, respectively, were used. Blood sampling was performed by vein‐puncture and indwelling catheters, respectively, of both ulnaris vein and artery of animals which had been fasting for 12 hours. Determinations of ionized calcium concentration was carried out by use of an ion‐selective electrode (ICA1), and of total calcium concentration in plasma using atomic absorption, respectively. After attainment of sexual maturity total calcium in roosters rose from 2.73 to 2.85 mmol/l, whereas ionized calcium fell from 1.45 to 1.40 mmol/l and per cent fraction of ionized/total calcium (extent of ionization) from 53.1 to 49.1%, respectively. After attainment of sexual maturity in hens, the content of total calcium in venous blood rose from 2.78 to 5.78 mmol/l, and of ionized calcium from 1.45 to 1.59 mmol/l, respectively, whereas the extent of ionization was significantly reduced from 52.2 to 27.1%. On comparison of the vein‐puncture and indwelling catheter technique a marked influence of the blood sampling procedure on calcium concentrations measured in blood of chicken became evident. On attainment of sexual maturity both the arterial and the venous blood (sampled from indwelling catheters) of hens showed levels of ionized calcium about 8% higher as compared to roosters. The extent of ionization amounted to 39% in sexually mature females as opposed to 51% in males. Concentrations of calcium ions in venous blood averaged 0.07 to 0.08 mmol/l higher compared to arterial blood in either sex. Concentration of ionized calcium did not exhibit any correlation to total calcium in adult animals. However, the concentration of ionized calcium was inversely correlated to blood pH. Zusammenfassung Ziel der vorliegenden Arbeit war es, das Verhalten der Calciumionenkonzentration und die Korrelation zwischen dem Gesamtcalcium und dem ionisierten Calcium im Blut in Abhängigkeit von Alter und Geschlecht beim Haushuhn zu untersuchen. Gleichzeitig sollte dabei der Einfluß der Blutentnahmetechnik auf die Meßparameter mit erfaßt werden. Als Versuchstiere dientèn männliche und weibliche Leghorntiere der Altersgruppen 3, 9, 25 und 80 Wochen. Die Blutentnahme erfolgte mittels Punktion bzw. Verweilkatheter aus der A. und V. ulnaris bei 12stündiger Nüchternheit der Tiere. Die Bestimmung des ionisierten Calciums erfolgte mit ionenselektiver Elektrode (ICA1) und die des Gesamtcalciums im Plasma mittels Atomabsorption. Die Untersuchungen ergaben, daß bei männlichen Tieren bis nach dem Eintritt der Geschlechtsreife das Gesamtcalcium von 2,73 auf 2,85 mmol/l ansteigt, der ionisierte Calciumspiegel von 1,45 auf 1,40 mmol/l und der prozentuale A...
