The protective effect of α-hederin, the active constituent of Nigella sativa, on tracheal responsiveness and lung inflammation in ovalbumin-sensitized guinea pigs

Abstract: Many investigations have demonstrated the prophylactic effect of Nigella sativa on asthma disease. One of its active constituents is α-hederin. In the present study, the preventive effect of two different concentrations of α-hederin on tracheal responsiveness and lung inflammation in ovalbumin-sensitized guinea pigs was examined. Forty male adult Dunkin-Hartley guinea pigs were randomly divided into the control (C), sensitized (S) and sensitized pretreated groups with thymoquinone (3 mg/kg i.p., S + TQ), low-d… Show more

“…NS and its active compounds thymoquinone, nigellone and α-hederin have been investigated in eighteen whole or cellular animal models and one human cellular model related to asthma. NS oil, thymoquinone or α-hederin showed anti-inflammatory and immunomodulatory effects in seven studies (Abbas et al, 2005, Balaha et al, 2012, El Gazzar et al, 2006, Mansour and Tornhamre, 2004, Saadat et al, 2015, Saleh et al, 2012, Shahzad et al, 2009). NS extracts, thymoquinone or α-hederin demonstrated a bronchodilatory or relaxant effect in six studies (Abd El Aziz et al, 2011, Al-Majed et al, 2001, Boskabady et al, 2008, Gilani et al, 2001, Keyhanmanesh et al, 2013, Saadat et al, 2015).…”

“…In addition, immune pathways driven by T-helper 2 (Th 2 ) cells which produce IL-4, IL-5, and IL-13 are contributing in the pathophysiology of this disease. The imbalance of Th 1 /Th 2 imbalance toward increased Th 2 activity was indicated in the pathogenesis of asthma and allergies [3, 4]. Cytokines released from Th 2 cells like IL- 4 and IL- 5 also stimulate IgE production by B cells which plays a role in allergic asthma [5].…”

“…In the present study, a-hederin increased levels of miRNA-133a mRNA expression, which indicates the possible therapeutic effect of a-hederin on sensitized rats. In previous studies, the preventive effect of a-hederin on tracheal responsiveness, blood cytokines and lung inflammation of sensitized guinea pigs was demonstrated Saadat et al, 2015]. Moreover, a-hederin reduced the levels of miRNA-126, IL-13 mRNA and lung pathological changes in comparison with OVA-sensitized rats [Fallahi et al, 2016].…”
