Effect of α‐Hederin on IL‐2 and IL‐17 mRNA and miRNA‐133a Levels in Lungs of Ovalbumin‐Sensitized Male Rats

Abstract: α-hederin, a saponin that is a major constituent of English Ivy (Hedera helix) is effective in the treatment of asthma. In the present study, the effect of α-hederin on lung tissue pathology and the levels of the inflammatory mediators; IL-2 mRNA, IL-17 mRNA, and MicroRNAs (miRNA)-133a was evaluated in a rat ovalbumin (OVA)-sensitized model of asthma. Rats were divided randomly into control (C), OVA-sensitized (S), OVA-sensitized pretreated with the antioxidant, thymoquinone (3 mg/kg, S + TQ) or OVA-sensitized… Show more

“…Similarly to hederagenin molecule (hederagenin is the aglycone part of numerous saponins, e.g. α-hederin) beneficial effect of α-hederin (a water-soluble pentacyclic triterpenoid saponin) in ovalbumin-sensitized rats was described, suggesting that α-hederin affects the IL-17 secretion pathways, altering miRNA-133a expression (Ebrahimi et al 2016). To support our hypothesis of the possible effect of terpenes, it was described, that synthetic triterpenoids also inhibit IL-17 and could improve autoimmune disease in mice (Fitzpatrick et al 2014).…”

“…In the current study, the paracrine effects of intravenously injected rBMMSCs and relevant CM phase were individually investigated in terms of the expression of IL-4, IL-13, IL-10, miRNA133, miRNA155 along with pathological changes in the lungs of sensitized rats. Our pathological findings revealed that lesions observed in the lung tissue of the ovalbumin-sensitized groups were significantly higher than the control rats, indicating the rat model of asthma was constructed successfully (Ebrahimi et al 2016;Mohammadian et al 2016b). Similar to previous findings, a significant reduction in the expression of IL-10 and miRNA133, coincided with a remarkable increase in the expression of IL-4, IL-13 and miRNA155 were seen in all sensitized groups as compared with healthy rats (Rankin et al 1996;Chiba et al 2009;Dong et al 2014;Malmhäll et al 2014;.…”

“…Moreover, antiischemic effects of black cumin seed, oil, and its active constituents in different organs including brain, kidneys, heart, liver, and intestine have been reported owing to their immunomodulatory and antioxidant properties (Oskouei, Akaberi, & Hosseinzadeh, 2018). Administration of α‐hederin (0.02 mg/kg) in ovalbumin (OVA)‐sensitized rats as asthma model affected the interleukin (IL)‐2 and IL‐17 secretion pathways, altering miRNA‐133a expression (Ebrahimi et al, 2016).…”
