The protective effect of melatonin supplementation against taxol-induced testicular cytotoxicity in adult rats

Abstract: The aim of the present investigation was to study the toxic influences of taxol (TXL) on the testes of rats and the protective impact of melatonin (MLT) against such effects. Rats were classified into control, sham, TXL, MLT, and MLT+TXL-treated groups. Histological and ultrastructural changes were observed in testicular tissues of TXL-intoxicated rats including thickening of tunica albuginea and degenerative alterations in spermatogenic, Sertoli, and Leydig cells. A significant increase (Pp0.05) was found in … Show more

“…The present results revealed a remarkable serum sex hormone levels improvement in MLT-treated irradiated rats, thereby restoring germ-cell and somatic cell population in the testis. These findings are compatible with a recent report that MLT directly affects the testis and testosterone synthesis from Leydig cells in animals [ 56 ]. Thus, MLT may promote male reproductive performance and increase testosterone synthesis in mammalian Leydig cells.…”

“…But it has severe side effects in other organs, including the reproductive system (Honore et al, 2005;Wang et al, 2017). Many histological and ultrastructural changes, including thickening of the tunica albuginea and basal lamina, and degenerative changes in spermatogenic, Sertoli and Leydig cells were seen in the testis of rats treated with Taxol, but co-treatment of melatonin and taxol caused the modulation of histomorphometric and ultrastructural changes, and with increased expression of BCL-2 and decreased expression of P53 and Caspase 3, decreased the amount of apoptosis in testicular tissue (Aboelwafa et al, 2022).…”

“…Quercetin co-treatment showed a significant reduction in the interstitial space in the arsenic-treated group [ 78 ]. Melatonin co-treatment showed a significant reduction in the thickness of tunica albuginea in the taxol-treated group [ 79 ].…”
